Warfarin and flax seed oil interaction and a mechanism for Polyunsaturated Fatty Acid and Warfarin interactions

چکیده
Warfarin is the most widely prescribed oral anticoagulant in North America and the 11th most prescribed drug in the United States; however, it is underutilized in those who meet criteria for warfarin therapy due to concerns regarding bleeding complications. Warfarin therapy requires frequent monitoring via the International Normalized Ratio (INR) which places considerable time and financial burdens on our healthcare system. In addition, a plethora of genetic, food, drug and herbal interactions are associated with warfarin use. In a recent systematic review of warfarin interactions, 187 separate reports were investigated and only 14 were deemed 'highly probable' for potentiation of warfarin's effect on the INR. One of these 14, fish oil, has recently been touted as a valuable source of omega-3 polyunsaturated fatty acids (PUFAs). Recent data suggest that omega-3 PUFAs like those found in fish oil and flax seed oil (a plant source of omega3 PUFAs) play a protective role cardiovascular disease. Many patients with cardiovascular disease have the potential to receive concomitant warfarin and omega-3s. However, only interactions with warfarin and fish oil have been reported. We present the first such observed ‘possible’ interaction, as assessed by the Drug Interaction Probability Scale (DIPS), and propose a novel mechanism to explain the observed interaction in not only flax seed oil, but fish oil as well.

Flax (Linum usitatissimum L
The archaeological evidence of flax cultivation dates back to >6000 BC and it is considered as one of the oldest and most useful crops. Components of flax have diverse uses. Cultivar development of flax is currently focused on enhancing the oil content and nutritional value to meet the demand of nutraceutical market supply, as an alternate source of fish oil, a rich source of eicosapentaenoic a...
متن کاملHeterologous expression of flax PHOSPHOLIPID:DIACYLGLYCEROL CHOLINEPHOSPHOTRANSFERASE (PDCT) increases polyunsaturated fatty acid content in yeast and Arabidopsis seeds
BACKGROUND Flax (Linum usitatissimum L.) is an agriculturally important crop with seed oil enriched in α-linolenic acid (18:3 (cisΔ9, 12, 15); ALA). This polyunsaturated fatty acid (PUFA) is the major determinant for the quality of flax seed oil in food, nutraceuticals and industrial applications. The recently identified enzyme: phosphatidylcholine diacylglycerol cholinephosphotransferase (PDCT...

Enrichment of tissues with ≥20-carbon n-3 PUFA like EPA is associated with positive cardiovascular outcomes. Stearidonic acid (SDA; 18 : 4n-3) and α-linolenic acid (ALA; 18 : 3n-3) are plant-derived dietary n-3 PUFA; however, direct comparisons of their impact on tissue n-3 PUFA content are lacking. Ahiflower(®) oil extracted from Buglossoides arvensis seeds is the richest known non-genetically...
متن کاملNatural phenolics greatly increase flax (Linum usitatissimum) oil stability
BACKGROUND Flaxseed oil is characterized by high content of essential polyunsaturated fatty acids (PUFA) promoted as a human dietary supplement protecting against atherosclerosis. The disadvantage of the high PUFA content in flax oil is high susceptibility to oxidation, which can result in carcinogenic compound formation. Linola flax cultivar is characterized by high linoleic acid content in co...
متن کاملProducing Omega-3 Enriched Eggs
As with traditional feed grains, the form in which flax seed is fed impacts egg production and hen performance (Choi et al. 1986, Vilarino et al. 1996). Flax seed form also affects the amount of omega-3 polyunsaturated fatty acid (PUFA) incorporated into the egg. Differences in the omega-3 content of eggs from laying hens fed whole versus milled flax seed have been reported, with many researche...
