Immunocytochemical labelling of haematological samples using monoclonal antibodies.

چکیده
Monoclonal antibodies have become increasingly important in the diagnosis and classification of neoplastic blood disorders. Immunofluorescent labelling of cell surface antigens has been the conventional method used for this purpose. However, this method of antigen detection presents a number of drawbacks when used in a routine hospital haematology laboratory: namely that blood or bone marrow samples must be processed within a short time of collection; antigen labelling is not permanent; and cell morphology cannot be visualised. The present paper describes the method for labelling routine haematological samples by an immunoalkaline phosphatase technique which is used routinely in this laboratory for leukaemia phenotyping. This approach has several practical advantages over conventional immunofluorescent labelling, i.e. staining can be performed on routinely prepared bone marrow and blood smears, samples can be stored for long periods before labelling, cell morphology can be visualised simultaneously with the antigen label and the staining reaction is permanent. As detailed below a wide range of cellular antigens can be detected in marrow and blood smears using this technique.

Immunocytochemical staining with monoclonal antibodies in cytologically "negative" serous effusions from patients with malignant disease.
In order to assess the practical value of immunocytochemical techniques in routine cytological diagnosis, a panel of three monoclonal antibodies (anti-CEA, Ca 1 and HMFG-2) were used to search for malignant cells in 53 samples of pleural or peritoneal fluid from 41 selected patients. All these patients suffered from malignant disease, but neoplastic cells had not been reported on conventional c...
متن کاملIdentification of Leishmania species using monoclonal antibodies in Isfahan
Background: Isfahan is one of the major foci of cutaneous leishmaniasis in Iran. According to the epidemiological studies, both urban and rural forms have been reported in Isfahan. Accurate identification and characterization of Leishmania is essential for the control of the disease. Several different molecular techniques for Leishmania identification are developed including use of monoclonal a...
متن کاملMonoclonal and Polyclonal Antibodies Specific to Human Fibromodulin
Background: The unique expression of fibromodulin (FMOD) in patients with chronic lymphocytic leukemia (CLL) has been previously reported. Detecting FMOD in CLL patients using specific anti-FMOD mAbs might provide a promising method in detection, monitoring, and prognosis of CLL. Objectives: In this study, we aimed for producing specific antibodies agains...

In the current study, immunocytochemical typing of intermediate filaments was used for a differential diagnosis of human lung tumors from transthoracic fine-needle aspiration biopsies (TFNAB). The authors have compared the cytologic diagnosis of 53 lung cancer cases with the immunofluorescence patterns obtained using a panel of monoclonal antibodies, five of which (KG 8.13, KM 4.62, Ks B.17, KS...
متن کاملMONOCLONAL ANTIBODIES TO HUMAN ALPHA-FETOPROTEIN
Alpha-fetoprotein (AFP), a serum glycoprotein belonging to the onco-developmental proteins group, serves as a marker both in cancer research and in studies concerning fetal development and fetal pathophysiology. Monoclonal anti-AFP antibodies are essential reagents in developing appropriate techniques for measurement of this protein. In this study, in order to produce anti-AFP monoclonal a...
