Microsatellite primers for genotyping common marmosets (Callithrix jacchus) and other callitrichids.

uct was obtained under the same PCR conditions as those used for A. argenteus. However, three primer sets (MSAA-3, MSAA-4 and MSAA-6) gave PCR products in the amplification using low annealing temperatures at 45 ¡C, 35 ¡C and 40 ¡C, respectively, although the remaining sets did not provide any products even at the low annealing temperatures (e.g. 10 ¡C below the annealing temperature in Table 1). Each of the three loci also showed an allelic variation in A. speciosus, i.e. MSAA-3 with six alleles (121Ð135 bp), MSAA-4 with six alleles (214Ð226 bp) and MSAA-6 with four alleles (234Ð244 bp), respectively. Several recent studies reported nonamplifying (or null) microsatellite alleles in mammals, including humans (Callen et al. 1993), deer (Pemberton et al. 1995), bears (Paetkau & Strobeck 1995) and grey-sided voles (Ishibashi et al. 1996). In the present study, the observed heterozygosities at the MSAA-2 and MSAA-5 loci differed significantly from the state of HWE, the former showing a greater difference than the latter (Table 1). These loci may have null alleles in the examined population of A. argenteus. Although these loci may need a revision for the primer sequences, especially for MSAA-2, the remaining four microsatellite loci will become a potent DNA marker for investigating the genetic variation in A. argenteus.
