چکیده

Background Median survival of advanced malignant pleural mesothelioma (MPM) is less than 2 years, and novel treatments are urgently needed. Immunotherapy with adoptive T cell transfer is an attractive approach that could be added to current therapeutic concepts. Fibroblast activation protein (FAP) is expressed by all subtypes of MPM [1] and can serve as target of re-directed T cells harboring an anti-FAP chimeric antigen receptor (CAR) leading to site-specific T cell activation.

Pre-clinical testing of malignant fibroblast activation protein-specific re-directed T cells for treatment of pleural mesothelioma

Introduction Malignant pleural mesothelioma (MPM) is an incurable malignant disease that results mostly from chronic exposition to asbestos. Fibroblast activation protein (FAP) is predominantly expressed on the surface of reactive tumor-associated fibroblasts and on particular cancer types. Therefore, FAP is an attractive target for adoptive T cell therapy. T cells can be re-directed by gene tr...

The Economic Impact of Clinical Research in an Italian Public Hospital: The Malignant Pleural Mesothelioma Case Study

Background The current economic constraints cause hospital management to use the available public resources as rationally as possible. At the same time, there is the necessity to improve current scientific knowledge. This is even more relevant in the case of patients with malignant pleural mesothelioma (MPM), given the severity of the disease, its dismal prognosis, and the cost of chemotherapy ...

The Role of Immunohistochemistry Studies in Distinguishing Malignant Mesothelioma from Metastatic Lung Carcinoma in Malignant Pleural Effusion

Background and Objective: Early diagnosis of malignant pleural mesothelioma (MPM) is the key point of its treatment. The main problem is the precise diagnosis of mesothelioma and its differentiation from metastatic lung adenocarcinoma. Mesothelioma exhibits complex immunohistochemical characteristics. The aim of this study was to study hybrid immunohistochemistry in the differe...

Pemetrexed Maintenance Therapy Following Bevacizumab-Containing First-Line Chemotherapy in Advanced Malignant Pleural Mesothelioma

Malignant pleural mesothelioma (MPM) is a lethal disease with poor prognosis. The combination of cisplatin and pemetrexed has been confirmed as the standard of care for nonoperable MPM. Data have shown that the adoption of pemetrexed maintenance therapy (PMT) following first-line treatment appears extremely promising. We describe a 57-year-old man diagnosed as advanced MPM. We treated this patie...

A phase 2 randomized, double-blind, placebo-controlled study of tremelimumab for second and third line treatment in patients with unresectable pleural or peritoneal mesothelioma

Introduction Malignant mesothelioma (MM) is an uncommon cancer, caused principally by asbestos exposure. No treatments after first-line platinum-pemetrexed [1] have shown survival benefit [2], thus novel approaches are needed. Asbestos exposure induces immunosuppression and immune dysfunction in the mesothelium environment, mainly by hyperactivation of regulatory T lymphocytes and overproductio...
