چکیده
The measurement of the torque-speed curve is usually done by fixing the cell to a glass slide and tethering a polystyrene bead to the flagellar hook. An optical trap monitor the rotational speed of the bead and the motor torque is calculated from τ = (ξL + ξR)ω ≈ ξLω, where ξR is the drag coefficient due to the internal friction in the motor, ξL is the bead drag coefficient, and ω is the angular velocity. The torque-speed curve is then obtained changing ξL by varying the bed size [1] or the viscosity of the external medium. An alternative method is to tether a cell to a glass coverslip by one of its shortened flagellar filament and expose the cell to a rotating electric field [2]. Then the motor is broken spinning the cell backward. The difference between the cell body speeds before and after the broke of the motor, at the same value for the external applied torque, is proportional to the motor torque.

Dynamics of the bacterial flagellar motor with multiple stators.
The bacterial flagellar motor drives the rotation of flagellar filaments and enables many species of bacteria to swim. Torque is generated by interaction of stator units, anchored to the peptidoglycan cell wall, with the rotor. Recent experiments [Yuan J, Berg HC (2008) Proc Natl Acad Sci USA 105:1182-1185] show that at near-zero load the speed of the motor is independent of the number of stato...

UNLABELLED It is becoming clear that the bacterial flagellar motor output is important not only for bacterial locomotion but also for mediating the transition from liquid to surface living. The output of the flagellar motor changes with the mechanical load placed on it by the external environment: at a higher load, the motor runs more slowly and produces higher torque. Here we show that the num...
متن کاملThe Limiting Speed of the Bacterial Flagellar Motor.
Recent experiments on the bacterial flagellar motor have shown that the structure of this nanomachine, which drives locomotion in a wide range of bacterial species, is more dynamic than previously believed. Specifically, the number of active torque-generating complexes (stators) was shown to vary across applied loads. This finding brings under scrutiny the experimental evidence reporting that l...
متن کاملExchange of rotor components in functioning bacterial flagellar motor.
The bacterial flagellar motor is a rotary motor driven by the electrochemical potential of a coupling ion. The interaction between a rotor and stator units is thought to generate torque. The overall structure of flagellar motor has been thought to be static, however, it was recently proved that stators are exchanged in a rotating motor. Understanding the dynamics of rotor components in function...
متن کاملThe C-terminal periplasmic domain of MotB is responsible for load-dependent control of the number of stators of the bacterial flagellar motor
The bacterial flagellar motor is made of a rotor and stators. In Salmonella it is thought that about a dozen MotA/B complexes are anchored to the peptidoglycan layer around the motor through the C-terminal peptidoglycan-binding domain of MotB to become active stators as well as proton channels. MotB consists of 309 residues, forming a single transmembrane helix (30-50), a stalk (51-100) and a C...
