Development and regulation of ketogenesis in isolated hepatocytes from newborn rabbits.

چکیده
Physiological hyperketonemia develops after birth in most mammals (Girard and Ferré, 1982). Since the body lipid stores in newborns are usually low, ketone body precursors are supplied by milk which is a high-fat diet. However, in a few species like rabbits, the fetus is endowed with large hepatic lipid stores and starved newborns show an increase in blood ketone body concentrations 6 to 12 h after birth (Callikan et al., 1979). The aim of the present study was to

Induction of ketogenesis and fatty acid oxidation by glucagon and cyclic AMP in cultured hepatocytes from rabbit fetuses
The effects of pancreatic hormones and cyclic AMP on the induction of ketogenesis and long-chain fatty acid oxidation were studied in primary cultures of hepatocytes from fetal and newborn rabbits. Hepatocytes were cultivated during 4 days in the presence of glucagon (106 M), forskolin (2 x 10'M), dibutyryl cyclic AMP (10-' M), 8-bromo cyclic AMP (10-' M) or insulin (10-' M). Ketogenesis and fa...
متن کاملMetabolic fate of non-esterified fatty acids in isolated hepatocytes from newborn and young pigs. Evidence for a limited capacity for oxidation and increased capacity for esterification.
In hepatocytes isolated from 48 h-old starved of suckling newborn pigs or from 15-day-old starved piglets, the rate of ketogenesis from oleate or from octanoate is very low. This is not due to an inappropriate fatty acid uptake by the isolated liver cells, but results from a limited capacity for fatty acid oxidation. Some 80-95% of oleate taken up is converted into esterified fats, whatever the...
متن کاملRelationship between ketogenesis and gluconeogenesis in isolated hepatocytes from newborn rats.
In hepatocytes from 1-day-old rats, active gluconeogenesis occurs in parallel with active ketogenesis, although the carbon atoms of non-esterified fatty acids do not participate in glucose synthesis. Once a significant ketogenesis is established, a further increase does not enhance gluconeogenesis. Indeed, octanoate is more ketogenic than oleate, but stimulates gluconeogenesis to a similar extent.
متن کاملSuckling-Weanling Transition in the Rat STUDIES WITH ISOLATED HEPATOCYTES By MANUEL BENITO, EMMA WHITELAW* and DERMOT
The rates of ketogenesis from endogenous substrates, butyrate or oleate, have been measured in isolated hepatocytes from suckling and weanling rats. Ketogenesis from endogenous substrate and from oleate decreased on weaning, whereas the rate from butyrate remained unchanged. It is concluded that the major site of regulation of ketogenesis during this period of development involves the disposal ...

Thus in a system in uitro using rat adipose tissue, the addition of H D lipoprotein leads to an inhibition of lipolysis of ‘Intralipid’. This phenomenon could be the effect of competition between H D boprotein and substrate for enzyme association, perhaps through its apoprotein CII component, which can bind to lipoprotein b a s e . From our results the Percentage inhibition was greatest at the ...
