A UCL Some Studies of Artificial Enzyme Systems
نویسنده
چکیده
This thesis describes a novel approach to the rational design o f artificial esterases and aldolases. The Introduction provides a literature summary of the previous approaches that have been employed towards the design and synthesis of artificial enzyme systems. Chapter 2 describes the preparation and reactivity of a number of polymer based artificial enzymes, which are capable of catalysing ester hydrolysis. The study has involved the incorporation of a histidine catalytic group together with specifically designed peptide binding groups within a polymeric backbone. The binding groups were specifically selected according to their binding affinity towards an appropriate transition state analogue. The synthesis of peptide binding sites and thus incorporation of these, together with the histidine catalytic group into a polymer backbone, using standard peptide chemistry has been outlined. The results to an investigation of the influence of different pH, solvent and substrate concentration on the activity of artificial esterases are presented. Chapter 3 describes preliminary work undertaken towards the design and synthesis of artificial aldol catalysts. The aldolases, which feature a proline residue attached to a polymer backbone are shown to selectively catalyse aldol reactions using aromatic aldehydes as electrophilic partner. Chapter 4 describes the detailed experimental procedures used.

Design and Docking Study of Some Pyrimidine derivatives as Antimalarial Agents
Background and Aim: According to the latest estimate published by the World Health Organization in 2017, there are 219 million malaria cases and 435,000 deaths. With the emergence of drug-resistant strains in malaria, there is a need for new drug targets every time. In this study, the design and docking study of the pyrimidine derivatives for inhibiting Methionine aminopeptidase1B enzyme (Metap...
متن کاملUCL Depthmap 7: From Isovist Analysis to Generic Spatial Network Analysis
This short paper discusses some of the new functions available in UCL Depthmap 7. UCL Depthmap’s original purpose was to perform integration analysis of isovists just as the integration of axial lines could be calculated. It has now developed in a number of different directions, so it has become closer to a traditional GIS system with analysis functions for graphs built in. In doing so, it has ...
متن کاملUCL-GLORP - An ORM for Common Lisp
UCL-GLORP is a Common Lisp implementation and extension of GLORP (Generic Lightweight Object-Relational Persistence), an Object-Relational Mapper for the Smalltalk language. UCL-GLORP is now a mature framework that largely extends GLORP and that takes advantage of some of Common Lisp unique features. This paper illustrates UCL-GLORP and discusses some of the challenges that we faced in order to...
متن کاملOn the convergence speed of artificial neural networks in the solving of linear systems
Artificial neural networks have the advantages such as learning, adaptation, fault-tolerance, parallelism and generalization. This paper is a scrutiny on the application of diverse learning methods in speed of convergence in neural networks. For this aim, first we introduce a perceptron method based on artificial neural networks which has been applied for solving a non-singula...
