چکیده
The development of nanocarriers that selectively release guest molecules on sensing a particular biological signal is being actively pursued in nanomedicine for diagnostic and therapeutic purposes. Here we report a protein-based nanocarrier that opens in the presence of intracellular adenosine-5'-triphosphate (ATP). The nanocarrier consists of multiple barrel-shaped chaperonin units assembled through coordination with Mg(2+) into a tubular structure that protects guest molecules against biological degradation. When its surface is functionalized with a boronic acid derivative, the nanocarrier is able to enter cells. The hydrolysis of intracellular ATP into adenosine-5'-diphosphate (ADP) induces conformational changes of the chaperonin units, which in turns generate a mechanical force that leads to the disassembly of the tube and release of the guests. This scission occurs with a sigmoidal dependence on ATP concentration, which means that the nanocarrier can differentiate biological environments in terms of the concentration of ATP for selective guest release. Furthermore, biodistribution tests reveal preferential accumulation of the nanocarriers in a tumour tissue.

A mechanochemical theory for the ATP-fuelled biomolecular motors
Biomolecular motors are normally single or complex biomolecules exerting mechanical forces over molecular and cellular scales. The ATP-fuelled biomolecular motors can transduce the chemical energy from ATP hydrolysis into forces and motions in cells. In biomolecular motors, transport reactions are both stoichiometric and enzymatic. We outline a mechanochemical theoretical framework for biomolec...
متن کاملA cyclic RGD-coated peptide nanoribbon as a selective intracellular nanocarrier.
We have synthesized a peptide-based supramolecular building block consisting of a cyclic Arg-Gly-Asp (cRGD) peptide segment and a beta-sheet-forming peptide segment. The block peptide was shown to self-assemble into a cRGD-coated nanoribbon structure, as revealed by circular dichroism (CD), dynamic light scattering (DLS), and transmission electron microscopy (TEM) studies. We have shown that th...
متن کاملHost-guest complexation driven dynamic supramolecular self-assembly.
Host-guest complexation between pillararene trimer 1 and biviologen 2 was used to fabricate the dynamic supramolecular self-assembly, which exhibits a reversible multidimensional transformation from 0D to 3D upon concentration changes. As a comparison, assemblies built by the complexation between 1,4-dimethoxypillar[5]arene and 2 only show spherical morphology under similar conditions.
متن کاملDual Space Control of a Deployable Cable Driven Robot: Wave Based Approach
Known for their lower costs and numerous applications, cable robots are an attractive research field in robotic community. However, considering the fact that they require an accurate installation procedure and calibration routine, they have not yet found their true place in real-world applications. This paper aims to propose a new controller strategy that requires no meticulous calibration and ...
متن کاملA cholic acid-based fluorescent chemosenor for the detection of ATP.
A novel ditopic cholic acid-based fluorescent chemosensor for ATP, 1a, was designed and synthesized. Its interactions with phosphates, AMP, ADP, ATP, CTP, GTP, and TTP have been investigated. When ATP was added to a 1:1 aqueous CH3CN solution of the sensor at pH 7.4, a significant decrease in fluorescence of 1a was observed, whereas other guest molecules showed a much smaller effect. The comple...
