چکیده

Surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) is a high throughput analytical technique capable of detecting low molecular weight analytes, including illicit drugs, and with potential applications in forensic toxicology as well as athlete and workplace testing, particularly for biological fluids (oral fluids, urine and blood). However, successful detection of illicit drugs using SALDI-MS often requires extraction steps to reduce the inherent complexity of biological fluids. Here, we demonstrate an all-in-one extraction and analytical system consisting of hydrophobically functionalized porous silicon microparticles (pSi-MPs) for affinity SALDI-MS of prescription and illicit drugs. This novel approach allows for the analysis of drugs from multiple biological fluids without sample preparation protocols. The effect of pSi-MP size, pore diameter, pore depth and functionalization on analytical performance is investigated. pSi-MPs were optimized for the rapid and high sensitivity detection of methadone, cocaine and 3,4-methylenedioxymethamphetamine (MDMA). This optimized system allowed extraction and detection of methadone from spiked saliva and clinical urine samples. Furthermore, by detecting oxycodone in additional clinical saliva and plasma samples, we were able to demonstrate the versatility of the pSi-MP SALDI-MS technique.

Laser Desorption/ionization Mass Spectrometry Using Tunable Nanoporous Structures

ABSTRACT The performance of porous silicon and cobalt glancing angle deposited (GLAD) thin films for matrix-free laser desorption/ionization-MS analysis of carbohydrates, peptides and drugs with molecular weights < 2,500 Dalton is presented. The performance of these films for selected peptides and small drugs detection is similar to the best reported for desorption/ionization on electrochemical...

Small-molecule analysis with silicon-nanoparticle-assisted laser desorption/ionization mass spectrometry.

Silicon nanopowder (5-50 nm) was applied as a matrix for the analysis of small molecules in laser desorption/ionization mass spectrometry. In contrast with conventional matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry, the matrix background interference in the low mass range was significantly reduced. Effects of the particle size and sample preparation proced...

Small Molecule Matrix - Free Laser Desorption / Ionization Mass Spectrometry

Modified Si wafers and nanoparticles (5-50 nm) have been developed and explored to assist laser desorption ionization mass spectrometry (LDI-MS) for small molecule analysis. DIOS (desorption/ionization on silicon) plates were prepared according to published protocols. DIOS was tested and compared with the optimized silicon nanoparticles derivatized by penterfluorophenylchlorosilane (PFP). SPALD...

There is considerable interest in the highly parallelized mass spectrometry analysis of complex sample mixtures without any time-consuming prepurification. Porous silicon-based laser desorption/ionization mass spectrometry (pSi LDI-MS) is enabling technology for such analysis. Previous studies have focused on pSi surface functionalization to enhance sensitivity of detection and engineer surface...

Secondary electrospray ionization ion mobility spectrometry/mass spectrometry of illicit drugs.

A secondary electrospray ionization (SESI) method was developed as a nonradioactive ionization source for ion mobility spectrometry (IMS). This SESI method relied on the gas-phase interaction between charged particles created by electrospray ionization (ESI) and neutral gaseous sample molecules. Mass spectrometry (MS) was used as the detection method after ion mobility separation for ion identi...
