Expression Level of the nm23 Gene in Clonal Populations of Metastatic Murine and Human Neoplasms1

چکیده
The purpose of this study was to determine whether nm23 steadystate mRNA expression levels correlate with metastatic potential of mouse K-1735 melanoma cells, human KM 12 colon cancer cells, and human SN12 renal cancer cells. Since neoplasms are heterogeneous and contain subpopulations of cells with different metastatic potentials, we analyzed multiple sets of nonmetastatic and metastatic clones isolated from each neoplasm. In addition, we also examined nine somatic cell hybrids produced by the fusion of nonmetastatic and metastatic K-1735 clones. In the mouse melanoma, we found heterogeneity in nm23-l steady-state expression levels among the clones and hybrids that did not correlate with their metastatic phenotype. Clones isolated from human colon or renal carcinomas expressed similar levels of nm23-HI regard less of metastatic potential in nude mice. All of the human tumor cells were heterozygous for the nm^J-///-$pecific allelic DNA fragments, with no allelic deletions or gross alterations detected. Since the failure of tumor cells to produce metastasis can be due to multiple deficiencies, these data stress the importance of using independent clones with dif ferent metastatic potentials for the analysis of gene regulation of this process.

Background & Aim: Although studies have shown that ibuprofen has anticancer effects on many cancer cells, the mechanism of the ibuprofen anticancer effect in cancer cells is not still well understood. The aim of this study was to investigate the effect of cytotoxic concentration of ibuprofen on the expression level of MMP-9 and NM23 genes in cervical cancer cells. Materials & Methods: During t...

متن کاملEvaluation of Cytotoxicity Activity and NM23 Gene Expression in T47D Breast Cancer Cell Line Treated with Glycyrrhiza glabra Extract
Glycyrrhiza is a genus of about 20 accepted species in the legume family (Fabaceae), with a distribution in Asia, Australia, Europe, and the Americas. Recently, about 30 species from the genus Glycyrrhiza have been found and used in traditional medicine for treating cancer. Different studies confirmed that down regulation of Non-metastatic protein (NM23), a metastasis suppressor gene is related...

متن کاملTwo isotypes of murine nm23/nucleoside diphosphate kinase, nm23-M1 and nm23-M2, are involved in metastatic suppression of a murine melanoma line.
A series of sublines of a murine melanoma B16 of C57BL/6 origin were established and examined regarding their metastatic capacity and expression of nm23. The number of pulmonary metastases developed by these sublines was inversely correlated with the expression of two isotypes of nm23, nm23-M1 and nm23-M2. The cDNAs of nm23-M1, nm23-M2, and a combination of both were transfected into the highly...
