چکیده

There are some differences in cytotoxic actions between local anesthetics. In a study performed using rat thymocytes, lidocaine increased intracellular Zn concentration whereas tetracaine decreased it. However, thymocytes are not subject to local anesthetics. This study was conducted to confirm the actions of tetracaine and lidocaine on neurons. Tetracaine and lidocaine changed intracellular Zn levels in rat cerebellar granule neurons, as had been observed in rat thymocytes. The changes in intracellular Zn levels by these local anesthetics were statistically significant, but small. The mechanism of lidocaine-induced increase in intracellular Zn levels in neurons was similar to that in rat thymocytes. The mechanism of tetracaine-induced decrease in intracellular Zn levels in neurons did not seem to be consistent with that in rat thymocytes. Further studies will be needed to see if the changes in intracellular Zn levels by local anesthetics correspond to differences in cytotoxic activities since their actions on intracellular Zn levels of neurons were weak.

Increase in intracellular Zn2+ concentration by thimerosal in rat thymocytes: intracellular Zn2+ release induced by oxidative stress.

Thimerosal (TMR), an ethylmercury-containing preservative in pharmaceutical products, was recently reported to increase intracellular Zn(2+) concentration. Therefore, some health concerns about the toxicity of TMR remain because of physiological and pathological roles of Zn(2+). To reveal the property of TMR-induced increase in intracellular Zn(2+) concentration, the effect of TMR on FluoZin-3 ...

Nitroprusside increases intracellular Zn concentration without affecting cellular thiol content: A model experiment using rat thymocytes and FluoZin-3

Nitric oxide (NO) is cytotoxic under some conditions although it has physiological roles. It is recently proposed that the cytotoxicity of NO is resulted from its interaction with glutathione and zinc. Since we have revealed that a decrease in cellular content of non-protein thiols, presumably glutathione, induces intracellular Zn release, there is a possibility that the cytotoxicity of nitropr...

Fluorescence imaging study of extracellular zinc at the hippocampal mossy fiber synapse.

Although synaptically released, vesicular Zn(2+) has been proposed to play a neuromodulatory or neuronal signaling role at the mossy fiber-CA3 synapse, Zn(2+) release remains controversial, especially when detected using fluorescent imaging. In the present study, we investigated synaptically released Zn(2+) at the mossy fiber (MF) synapse in rat hippocampal slices using three chemically distinc...

Different effects of local anesthetics on extracellular signal-regulated kinase phosphorylation in rat dorsal horn neurons.

Local anesthetics, which are widely known to be neuronal voltage-gated Na(+) channel blockers, also affect a variety of other ion channels, N-methyl-d-asparate (NMDA) receptors and α-amino-3-hydroxy-5-methyl-4-izoxazolepropionic acid (AMPA) receptors. Glutamate, which is released from presynaptic fibers, activates extracellular signal-regulated kinase (ERK) through NMDA and AMPA receptors in sp...

Nitric oxide (NO) is a unique diffusible molecule in the vascular and nervous systems. Endogenous NO can modulate neuronal physiology and pathology by participating in many signaling pathways. Zinc is an essential metal ion for the body. In neurons, most zinc is accumulated in different intracellular pools. Recently, zinc was shown to modulate different physiological functions by acting as a si...
