The Beatability-aiding Effect of Aspergillus niger Crude Cellulase on Bleached Simao Pine Kraft Pulp and its Mechanism of Action

چکیده
Bleached simao pine kraft pulp was treated with Aspergillus niger crude cellulase produced by submerged fermentation using the pulp as the carbon source. The effects of the cellulase on the pulp beatability and mechanical properties were studied. Fourier transform infrared spectroscopy (FT-IR) was performed to study the effect of cellulase treatment on the cellulose crystallinity index. The fiber morphology difference before and after treatment was also revealed by atomic force microscope observation. Compared to the control pulp, the beating time of the cellulase-treated pulp with the dosage of 7 u/g could be reduced from 360 s to 260 s under the same beating degree of 48 o SR, which indicates a savings of about 28% beating energy consumption. The cellulase treatment had negative impacts on the pulp’s mechanical properties. The cellulase preferentially adsorbed on the fine surface. The cellulose amorphous region was easier to treat with cellulase than the crystalline region. Atomic force microscope images demonstrated that the primary wall of fibers was peeled off and the S1 layer of fibers came to the surface after the cellulase treatment.

Effect of Cellulases and Xylanases on Refining Process and Kraft Pulp Properties
Samples of bleached kraft pine cellulosic pulp, either treated with an enzyme preparation (a Thermomyces lanuginosus xylanase, an Aspergillus sp. cellulase, and a multienzyme preparation NS-22086 containing both these activities) or untreated, were refined in a laboratory PFI mill. The treatment with cellulases contained in the last two preparations significantly improved the pulp's susceptibil...
متن کاملThe Role of Cellulose Binding Domains in the Adsorption of Cellulases onto Fibers and Its Effect on the Enzymatic Beating of Bleached Kraft Pulp
The adsorption of cellulases onto fibers may be one of the most important factors affecting the enzymatic reaction between cellulases and fibers. This study investigated the adsorption kinetics involved, using isothermal adsorption equations. Cellulose binding domains (CBDs) were isolated from a commercial cellulase, and their role in the adsorption and enzymatic reaction was evaluated. Approxi...
متن کاملDissolving Pulp (alpha-cellulose) from Corn Stalk by Kraft Process
Corn stalk can be used as an alternative non-wood raw material to produce cellulosic dissolving grade pulp. In this work, the potential of corn stalk in the production of dissolving pulp (alpha-cellulose) was investigated. It was collect ed from a corn stalk farm of Kermanshah, Iran. The sequences of experimental work were: water and acid prehydrolysis at 160˚C for 30min, followed by Kraft pulp...
متن کاملBiocatalytic activity of Aspergillus niger xylanase in paper pulp biobleaching
Xylanase is a hemicellulase enzyme that catalyses the hydrolysis of xylan to xylose which is widely used in processing of feed, pulp and paper. It is produced by many microorganisms especially filamentous fungi like Trichoderma and Aspergillus. A potential xylanolytic fungal isolate Aspergillus niger was isolated from forest soils of Tirumala, AP, India, and its crude enzyme was checked for its...
متن کاملBiobleaching Effects of Crude Xylanase from Streptomyces griseorubens LH-3 on Eucalyptus Kraft Pulp
In this work, a xylanase-producing strain, Streptomyces griseorubens LH-3, was cultured, and the crude xylanase was prepared. Analysis of its enzymatic properties revealed that the crude xylanase possessed good thermal stability at temperatures below 60 o C, exhibited a wide pH range from 4.0 to 9.0, and was cellulase-free. This crude enzyme was used to treat eucalyptus kraft pulp, and the rele...
