چکیده

The second thiamine-decomposing bacterium, discovered by Kimura et al. (1, 2) was named by the Vitamin B Research Committee Bacillus aneurino lyticus Kimura et Aoyama (BKA) (3). By studying the properties of the enzyme produced by this bacillus, it was found that the action mechanism of the enzyme is completely different from that of Bacillus thiaminolyticus Matsukawa et Misawa (BMM), shell-fish, fish and plant sources, which have been studied so far.

Pharmacologic properties of polyethylene glycol-modified Bacillus thiaminolyticus thiaminase I enzyme.

We have previously shown that the bacterial enzyme thiaminase 1 has antitumor activity. In an attempt to make thiaminase I a more effective pharmaceutical agent, we have modified it by adding polyethylene glycol (PEG) chains of various lengths. We were surprised to find that 5k-PEGylation eliminated thiaminase cytotoxic activity in all cell lines tested. Both native thiaminase and 5k-PEGylated ...

Structure of a eukaryotic thiaminase I.

Thiaminases, enzymes that cleave vitamin B1, are sporadically distributed among prokaryotes and eukaryotes. Thiaminase I enzymes catalyze the elimination of the thiazole ring moiety from thiamin through substitution of the methylene group with a nitrogenous base or sulfhydryl compound. In eukaryotic organisms, these enzymes are reported to have much higher molecular weights than their bacterial...

Thiaminase activities and thiamine content of Pteridium aquilinum, Equisetum ramosissimum, Malva parviflora, Pennisetum clandestinum and Medicago sativa.

Thiaminase type 1 and 2 activities and thiamine content of five plants were determined. Of these Pteridium aquilinum and Equisetum ramosissimum were found to have considerably more thiaminase activity and lower thiamine content than Malva parviflora, Pennisetum clandestinum and Medicago sativa.

Thiaminase activity of crucian carp Carassius carassius injected with a bacterial fish pathogen, Aeromonas salmonicida subsp. salmonicida.

Dietary thiaminase I is a cause of thiamine deficiency in animals. The physiological significance of thiaminase in the organisms containing this enzyme is not known, nor are the factors causing variation in their thiaminase activity. Tests were performed to evaluate the effect a pathogen might have on thiaminase activity in fish, when analyzed both with a cosubstrate added (CATA tests) and no c...

Isolation of the thiamine degradation products formed by shellfish thiaminase.

The breakdown of thiamine by the action of shellfish thiaminase was demon strated by Fujita et al. (1) to be a base exchange reaction. Thereafter, other type of thiaminase which decomposes thiamine into 2-methyl-4-amino-5-hydroxymethylpyrimidine (Pm-CH2OH) and 4-methyl-5-ƒÀ-hydroxyethylthiazole (Th) was discovered. Then, thiaminase was classified into a base exchange type (Thiaminase ‡T) and a ...
