Effects of Ions on Nematocysts Isolated from Acontia of the Sea Anemone Caluactis Tricolor by Different Methods

1. Differences in nematocyst isolating procedures significantly affected the discharge characteristics of the acontial nematocysts isolated from Calliactis tricolor. 2. Most of the nematocysts isolated in artificial sea water (ASW) discharged instantaneously in 5 m m o i r ' EGTA and to a lesser degree in distilled water (DW), but they did not discharge in Ca-free ASW or 50mmoir ' CaCl2. 3. Most of the nematocysts isolated in lmoll" sodium citrate discharged instantaneously when treated with any of the above solutions. 4. Nematocysts isolated in DW or 1 mol I" sucrose exhibited a modest degree of discharge in Smmoll" EGTA after a considerable latent period. 5. Some of the present results appear to be inconsistent with the hypothesis that removal of Ca from the intracapsular fluid causes nematocyst discharge.
