Statistical Analysis of Gel Electrophoresis Data

چکیده
Two-dimensional gel electrophoresis (2-DE) methods such as two-dimensional polyacrylamide gel electrophoresis (2D-PAGE; O’Farrell (1975)) and two-dimensional difference gel electrophoresis (2D-DIGE; Ünlü et al. (1997)) are popular techniques for protein separation because they allow researchers to characterize quantitative protein changes on a large scale. Thus, 2-DE is frequently used as an initial screening procedure whereby results obtained generate scientific ideas for study. These technologies revolutionized the field of proteomics and biomarker discovery in their ability to detect protein changes either in differential expression or modification (Huang et al., 2006; Rai & Chan, 2004; Wulfkuhle et al., 2003; Zhou et al., 2002). Further, they are attractive because of their resolving power and sensitivity. 2-DE analyses, however, require personnel with significant wet laboratory expertise and can be time-consuming, thus potentially limiting the sample size for gels.

Electrophoresis and chromatography of wheat proteins: available methods, and procedures for statistical evaluation of the data.
Analysis of gluten proteins from the wheat grain endosperm has long challenged the analytical chemist. Several hundred unique polypeptides are present, many in large polymers. This complexity, plus useful relationships of composition to genotype and quality, encouraged development and application of electrophoresis and chromatography for gluten analysis. We review the methods of polyacrylamide ...
متن کاملProposal for a Standard Representation of Two-Dimensional Gel Electrophoresis Data
The global analysis of proteins is now feasible due to improvements in techniques such as two-dimensional gel electrophoresis (2-DE), mass spectrometry, yeast two-hybrid systems and the development of bioinformatics applications. The experiments form the basis of proteomics, and present significant challenges in data analysis, storage and querying. We argue that a standard format for proteome d...
متن کاملA Simpler and Faster Version of Two-Dimensional Gel Electrophoresis Using Vertical, Mini SDS-PAGE Apparatus
We have modified one of the most useful methods of protein separation; namely, two dimensional gel electrophoresis (2-DE). This modified version of 2-DE is not only simpler and easier but also faster than all the currently available methods. In this method, isoelectric focusing is carried out in the first dimension using a vertical sodium dodecyl sulfate polyacrylamide gel electrop...
متن کاملMultivariate Statistical Tools for the Evaluation of Proteomic 2D-maps: Recent Achievements and Applications
Two dimensional polyacrylamide gel electrophoresis (2D-PAGE) maps represent an unavoidable tool in many fields connected with proteome research, such as development of new diagnostic assays or new drugs. Unfortunately the information contained in the maps is often so complex that its recognition and extraction usually requires complex statistical treatments. Statistics accompanies many phases o...
متن کاملDiscriminant Eigengels: a Statistical Approach to 2-d Gel Analysis
Two-dimensional (2-D) polyacrylamide gel electrophoresis (PAGE) images are widely used in the area of Proteomics. In this paper, a general method for extracting features from sets of PAGE images is presented. This combines the conventional approaches of Principal Components Analysis and Fisher’s Linear Discriminant Analysis, to provide a statistical approach to feature extraction. After a brief...
متن کاملStatistical Evaluation of Methods for the Analysis of Dynamic Protein Expres- Sion Data from a Tumor Study
• In this article, we analyze time dependent protein expression data obtained from a proteome study of a neuroblastoma cell line. Neuroblastoma are common solid tumors which occur in early childhood. The expression data was obtained by difference gel electrophoresis (DIGE). It is known that the clinical outcome of neuroblastoma depends on the activation of different neurotrophin receptors by th...
