Urea, urease, cyanate, and the sickling of hemoglobin S.

چکیده
Urea inglucose solutions has been advanced as a chemotherapeutic agent in sickle cell disease because it has been found effective both in reversing and in blocking sickling. It has been suggested recently that this beneficial action of urea may be the result of formation of cyanate from urea and subsequent carbamylation of beta S globin chains in the hemoglobin S molecule. In this paper, we show thatthe urea molecule per se interfereswith sickling. SS cellsinoxygen-free atmosphere, protected from sickling by ureasugar solutions, were sickled after urease was added. Urea blocks and reverses sickling;ureaprotected sickle cells become sickled after incubation with urease; these effects are reversible and immediate, and are apparent in the deoxygenated state. In contrast, cyanate blocks but does not reverse sickling;cyanate-treated cells are unaffected by urease; these effects are permanent and requiretime; and hemoglobin S must firstbe oxygenated before carbamylation willoccur. Evi. dently, urea and cyanate interferewith sickling by distinctlyseparate molecular mechanisms. Possible mechanisms for the action of urea are discussed.

Effects of cyanate and 2,3-diphosphoglycerate on sickling. Relationship to oxygenation.
Cyanate and 2,3-diphosphoglycerate (2,3-DPG) both influence the oxygen affinity of hemoglobin. The studies presented here concern the effects of these compounds on the sickling phenomenon. The inhibitory effect of cyanate on sickling is largely due to the fact that it increases the percentage of oxyhemoglobin S at a given oxygen tension. In addition, cyanate inhibits sickling by a mechanism tha...
متن کاملThe unexpected pathway to the creation of the HbA1c test and the discovery of AGE's.
This story begins at a social affair in New York for a group of haematologists. At that time I was an Assistant Professor at the Rockefeller University trying without success to isolate erythropoietin from theurine of patients with aplastic anaemia. One of the participants, Alan Schecter, mentioned that there had been a recent report of the successful treatment for sickle cell anaemia crisis in...
متن کاملUrea decomposition facilitated by a urease model complex: a theoretical investigation.
Density functional theory calculations were used to examine the role of the urease model complex [Ni2(bdptz)(micro-OH)(micro-H2O)(H2O)2](OTs)3(bdptz=1,4-bis(2,2'-dipyridylmethyl)-phthalazine; OTs=tosylate) in the degradation of urea. An elimination mechanism that converts urea to ammonium cyanate was investigated in detail. The lowest energy pathway involves urea coordination through the oxygen...
متن کاملSpot test for detection of sickling hemoglobin.
We describe a simple, noninstrumental procedure for detecting hemoglobin S in samples of blood on filter paper, with use of small volumes of reagents. The method depends on the differential solubilities of reduced sickling and nonsickling hemoglobin in phosphate solutions (1, 2) or Sickledex reagent’ and the apparent solubilizing effect of urea on reduced hemoglobin S (3). The color of paired p...
متن کاملUrease. Part II. The Mechanism of the Zymolysis of Urea.
Since this paper was submitted for publication, a paper has appeared on the mechanism of urease by Mack and Villars [1923]. In this paper the authors agree with the present writer in accepting the Werner formula for urea. They have detected and estimated cyanate in solutions of urea undergoing zymolysis, and also have shown that the cyanate rises to a maximum and falls again towards the end of ...
