چکیده
The fraudulent practice of cheap meat in costly meat is common nowadays. Therefore, there is need of precise identification of the meat species. We included in our study dog meat, as it is the most easily available cheap meat for adulteration. For specific identification of origin of meat animal species, we evaluated the common meat animal species including dog meat by PCR –RFLP technique. All the samples from all species resulted amplicons of 456 bp on PCR except in chicken we obtained 444 bp product. These amplicons further subjected to restriction digestion with five enzymes including AluI, HhaI, ApoI, BspTI and MboII, which resulted in speces specific restriction profiles which enable the identification. We were able to differenciate chicken and dog meat with AluI and MboII alone, whereas AluI, HhaI, ApoI, BspTI were specific to beef, buffalo meat, mutton and chevon respectively.

Establishment and application of a fluorescent polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method for identifying porcine, caprine, and bovine meats.
A method of fluorescent Polymerase Chain Reaction-restriction fragment length polymorphism (PCR-RFLP) was applied as an analytical and quantitative tool for meat identification. Following alignments of the nucleotide sequences, an oligonucleotide primer pair was designed to amplify the partial sequences within the 12S ribosomal RNA (12S rRNA) gene of mitochondrial DNA from porcine, caprine, and...
متن کامل[Species identification of animal hair present as a contaminant in food by PCR-APLP method].
A rapid, simple and inexpensive method was developed for identifying the species of animal hair present as a contaminant in food. A polymerase chain reaction-amplified product length polymorphism (PCR-APLP) assay was applied to identify hair from human and others (cat, dog, rabbit, rat and mouse) or livestock (pig, cattle, horse, sheep, goat and chicken). The PCR primers were designed to amplif...
متن کاملIdentification of blood meals in field captured sand flies by a PCR-RFLP approach based on cytochrome b gene.
Leishmaniasis is a vector-borne disease transmitted by phlebotomine sand flies. Information about blood meal preferences in sand flies is essential to understand the epidemiology of the disease to adopt control measures. In previous studies, a polymerase chain reaction (PCR) of 359bp fragment of the conserved gene cytochrome b (cyt b) and further sequencing were applied in the study of blood me...

Meat species specification is an area which needs specialized attention in food forensics. It is a vital field to ensure the food safety to the consumers and conserves the laws related to meat and meat products. The adulteration of inferior quality meat into superior quality meat is a common practice all over the world. DNA-Based techniques can easily solve the problems of vetro-legal or forens...
