Theoretical Analysis of Mode of Heptanol Action on Smooth Muscle Synaptic Potentials

چکیده
A theoretical analysis has been undertaken of the synaptic or "junction" potentials of syncytial smooth muscle in order to examine the most likely mode of action of a chemical, 1heptanol, that has profound effects on neurot"ission in this tissue. The smooth muscle syncytium of the mammalian vas deferens has been modelled as a bidomain electrical grid which allows simulation of the spontaneous and evoked excitatory junction potentiak (SEWS and eEJPs) observed experimentally in this organ. Two hypotheses of heptanol action raised by its effects on the electrical responses of the vas have been examined, (i) inhibition of cell-to-cell electrical coupling in smooth muscle; (ii) inhibition of the stimulationevoked release of neurot"itter from the autonomic nerve supply. We find that the range of experimental findings can be explained consistently on the basis of the second, but not the first, hypothesis. Our results thus cast doubt on the current belief that heptanol, at the concentrations used empirically, specifically uncouples smooth muscle cells from one another, and indicate a novel, previously unsuspected biophysical made of action which merits close scrutiny.

Heptanol but not fluoroacetate prevents the propagation of spreading depression in rat hippocampal slices.
We investigated whether heptanol and other long-chain alcohols that are known to block gap junctions interfere with the generation or the propagation of spreading depression (SD). Waves of SD were triggered by micro-injection of concentrated KCl solution in stratum (s.) radiatum of CA1 of rat hippocampal tissue slices. DC-coupled recordings of extracellular potential (V0) were made at the injec...
متن کاملEffects of ionic parameters on behavior of a skeletal muscle fiber model
All living cells have a membrane which separates inside the cell from it's outside. There is a potential difference between inside and outside of the cell. This potential difference will change during an action potential. It is quite common to peruse action potentials of skeletal muscle fibers with the Hodgkin-Huxley model. Since Hodgkin and Huxley summarized some controlling currents like inwa...

Introduction: Considering the importance of uterine contractions in uterus retraction and reducing post-partum hemorrhage and the current findings on the effect of the alcoholic extract of pomegranate seed on the uterine contractility, only few studies were made on this issue. In the present study the cumulative effect of the aqueous extract of pomegranate seed on the uterine smooth muscle c...

Propagation in cardiac muscle is faster in the longitudinal than in the transverse axis of the cells. Yet, as a result of the larger upstroke velocity of action potentials propagating transversely, it has been suggested that longitudinal propagation is more vulnerable to block. To study the relation between conduction velocity and maximal upstroke velocity (Vmax), as well as the time course of ...
متن کاملGABA-mediated membrane oscillations as coincidence detectors for enhancing synaptic efficacy in the developing hippocampus
Spontaneously occurring neuronal oscillations constitute a hallmark of developmental networks. They have been observed in the retina, neocortex, hippocampus, thalamus and spinal cord. In the immature hippocampus the so-called ‘giant depolarizing potentials’ (GDPs) are network-driven membrane oscillations characterized by recurrent membrane depolarization with superimposed fast action potentials...
