Title of Document: BUTANOL PRODUCTION FROM GLYCEROL BY Clostridium pasteurianum IN DEFINED CULTURE MEDIAA PHENOTYPIC APPROACH. David Leonardo Ramos Sanchez, Master of Science, 2009 Directed By: Associate Professor Nam Sun Wang Department of Chemical and Biomolecular Engineering ABSTRACT The fluctuations in oil prices have stimulated the production of renewable biofuels, in particular the production of bioethanol and biodiesel. The production of biodiesel has expanded almost six fold in the past years. The ten wt% of the biodiesel process results in crude glycerol. Once a valuable product, nowadays glycerol is considered a waste and a surplus material. Its current low price makes it an attractive substrate for a fermentation process. Molecular genetics have unveiled new insights about solvent production in Clostridia. It has been recognized that endospore development and solvent formation share a regulatory mechanism. The solvent production, particularly the butanol fermentation of glycerol by Clostridium pasteurianum was studied. Taking advantage of the characteristics of the sporulation phenotype, the study of the butanol fermentation was approached. A relation between spore formation and butanol production was found in C. pasteurianum by applying molecular genetics concepts.

Influence of nutritional and operational parameters on the production of butanol or 1,3-propanediol from glycerol by a mutant Clostridium pasteurianum.
Butanol and 1,3-propanediol (1,3-PDO) are simultaneously produced by Clostridium pasteurianum from glycerol. In this study, random chemical mutagenesis of C. pasteurianum DSM 525 was conducted to improve its tolerance to butanol. Selected nutritional and operational parameters were evaluated to identify strategies that favour the production of each metabolite. From those experiments, it was pos...

BACKGROUND Biodiesel production results in crude glycerol waste from the transesterification of fatty acids (10 % w/w). The solventogenic Clostridium pasteurianum, an anaerobic Firmicute, can produce butanol from glycerol as the sole carbon source. Coupling butanol fermentation with biodiesel production can improve the overall economic viability of biofuels. However, crude glycerol contains gro...
متن کاملTowards improved butanol production through targeted genetic modification of Clostridium pasteurianum
Declining fossil fuel reserves, coupled with environmental concerns over their continued extraction and exploitation have led to strenuous efforts to identify renewable routes to energy and fuels. One attractive option is to convert glycerol, a by-product of the biodiesel industry, into n-butanol, an industrially important chemical and potential liquid transportation fuel, using Clostridium pas...

Interspecies electron transfer is a common way to couple metabolic energy balances between different species in mixed culture consortia. Direct interspecies electron transfer (DIET) mechanism has been recently characterised with Geobacter species which couple the electron balance with other species through physical contacts. Using this mechanism could be an efficient and cost-effective way to d...
متن کاملA Review of Process-Design Challenges for Industrial Fermentation of Butanol from Crude Glycerol by Non-Biphasic Clostridium pasteurianum
Butanol, produced via traditional acetone-butanol-ethanol (ABE) fermentation, suffers from low yield and productivity. In this article, a non-ABE butanol production process is reviewed. Clostridium pasteurianum has a non-biphasic metabolism, alternatively producing 1,3-propanediol (PDO)-butanol-ethanol, referred to as PBE fermentation. This review discusses the advantages of PBE fermentation wi...
