Gradient gel electrophoresis of human plasma lipoproteins.

چکیده
A rapid and simple method for separating and identifying plasma lipoproteins is described. The technique utilizes the “molecular sieving” properties of continuous concave gradient electrophoresis on a slab of polyacrylamide (4 to 26%) gel. Gradient gel electrophoresis is particularly suitable for separating lipoproteins according to their molecular weight. The technique can be used to demonstrate the major Iipoprotein classes in normal plasma and to identify abnormalities of lipoprotein metabolism. Components of a sample are better resolved by gradient gel electrophoresis than by electrophoresis on paper or cellulose acetate. The results obtained compare favorably with those achieved by preparative ultracentrif ugation. The technique is easily and rapidly performed in a routine clinical laboratory.

Concentration gradient electrophoresis of plasma from patients with hyperbetalipoproteinemia.
A modification of the Pratt and Dangerfield technique of acrylamide gel concentration gradient electrophoresis has been used to study low density lipoproteins from fasting human beings. Prestained plasma from normal subjects and from patients with Fredrickson's type II hyperbetalipoproteinemia was electrophoresed in continuous gradient gel columns (approximately 3.5-8%). Reproducible low densit...
متن کاملPresence of multiple subpopulations of lipoproteins of intermediate density in normal subjects.
Analysis of human plasma by gradient gel electrophoresis (GGE) revealed multiple distinct bands in the size range between very low density (VLDL) and low density lipoproteins (LDL). GGE of intermediate density lipoproteins (IDL) from normal subjects consistently showed two major bands with particle diameters in the range of 275 to 300 A. The larger, usually predominant subspecies was designated...
متن کاملScreening for lipoprotein[a] elevations in plasma and assessment of size heterogeneity using gradient gel electrophoresis.
Plasma was screened for the presence of lipoprotein[a] using 2-16% nondenaturing, polyacrylamide gradient gel electrophoresis. Gels were scanned with a densitometer after staining with Sudan black B. Bands that migrated above low density lipoprotein bands were identified as lipoprotein[a] by immunoblotting with polyclonal and monoclonal antibodies to apolipoprotein[a]. Lipoprotein[a] was measur...
متن کاملDensity gradient characterization of the high density lipoproteins in cholesterol-fed hyper- and hyporesponding patas monkeys (Erythrocebus patas).
Diet-induced changes in high density lipoprotein (HDL) density and size were studied in patas monkeys. When the animals were switched from a moderate fat-low cholesterol diet to a high fat-high cholesterol (HFHC) diet, the plasma apoA-I levels increased initially in all of the animals. The apoA-I levels remained elevated in monkeys able to maintain their plasma cholesterol concentrations near b...

A bstract. Lipoprotein classes isolated from the plasma of two patients with apolipoprotein Al (apo Al) and apolipoprotein CIII (apo CIII) deficiency were characterized and compared with those of healthy, ageand sex-matched controls. The plasma triglyceride values for patients 1 and 2 were 31 and 51 mg/dl, respectively, and their cholesterol values were 130 and 122 mg/dl, respectively; the pati...
