Wasp Polydnavirus Polymorphism

چکیده

In this paper, we analyze the polymorphisms of parasitoid wasp polydnavirus. The polydnavius genome is normally dormant within the genome of the wasp, but is converted into particles that are injected into the infected host along with the wasp eggs. These particles contain genes which inhibit the immune system of the infected host, and are necessary for the eggs to mature. Within a population, these particles have varied mutation rates, which suggest the polydnavirus is actively avoiding the immune system of the host. Most mutations are single nucleotide polymorphisms occurring within genes. In addition, the mutation rates indicate high selected pressure on certain genes of the polydnavirus, suggesting they are essential to the polydnavirus for the wasp eggs to mature.

Quantitation of a Glyptapanteles indiensis polydnavirus gene expressed in parasitized host, Lymantria dispar, by real-time quantitative RT-PCR.

Glyptapanteles indiensis is a polydnavirus-carrying wasp that parasitizes early instar gypsy moth larvae. During oviposition, the wasp injects calyx fluid containing polydnavirus along with its eggs into the host. Within the host, expression of polydnavirus genes triggers a set of changes in host physiology, which are of critical importance for the survival of the wasp. In the present study, a ...

Polydnavirus infection inhibits synthesis of an insect plasma protein, arylphorin.

The wasp Campoletis sonorensis injects a segmented, double-stranded DNA polydnavirus (CsPDV) along with its egg during parasitization of Heliothis virescens larvae. After parasitization, CsPDV protects the wasp egg and larva by selectively disabling the host's cellular immune response. Other host physiological systems including growth and development are affected to the apparent benefit of the ...

Visualization of polydnavirus sequences in a parasitoid wasp chromosome.

Polydnaviruses, obligatorily associated with endoparasitoid wasps, are unique in that their segmented genome is composed of multiple double-stranded DNA circles. We present here the first cytological evidence that virus segments are integrated in the wasp genome, obtained by using in situ hybridization of virus probes with viral sequences in the chromosomes of a wasp from the braconid family of...

A polydnavirus-encoded protein of an endoparasitoid wasp is an immune suppressor.

The molecular mechanism by which polydnaviruses of endoparasitoid wasps disrupt cell-mediated encapsulation reactions of host insects is largely unknown. Here we show that a polydnavirus-encoded protein, produced from baculovirus and plasmid expression vectors, prevents cell surface exposure of lectin-binding sites and microparticle formation during immune stimulation of haemocytes. The inactiv...

Excision of the polydnavirus chromosomal integrated EP1 sequence of the parasitoid wasp Cotesia congregata (Braconidae, Microgastinae) at potential recombinase binding sites.

Cotesia congregata polydnavirus (CcPDV) is essential for successful parasitism of Manduca sexta larvae by the braconid wasp Cotesia congregata. To determine the molecular mechanisms for the vertical transmission of CcPDV in the wasps, we analysed the different forms of the virus sequences containing the gene encoding the early parasitism-specific protein 1 (EP1). By a detailed molecular analysi...
