Endosomes make tracks
نویسنده
چکیده
arly endosomes may help to create their own short-range tracks, according to a report from Stéphane Gasman, Yannis Kalaidzidis, and Marino Zerial (Max Planck Institute, Dresden, Germany). The results also suggest parallels between membrane trafficking, signaling, and regulation of the cytoskeleton. Trafficking of early endosomes requires the small GTPase RhoD. Zerial’s group identified the first known downstream effector for RhoD, hDia2C, a relative of the actin-polymerizing protein formin. In vivo, RhoD recruited hDia2C onto endosomes, where the two proteins probably stabilize the association of endosomes with the actin network, as their overexpression caused the vesicles to align along actin filaments. In the overexpressing cells, endosomal motility was also slowed and long-range trafficking was inhibited, probably because the endosomes were unable to jump from actin to microtubules, where Rab5 stimulates longrange movements. The results indicate a similarity between endosome motility and adhesion formation. Endosome-localized hDia2C recruited and activated c-Src kinase, which was required for the effects of RhoD and hDia2C on trafficking. Src is therefore another example of a signaling molecule required for organelle transport, although its relevant phosphorylation targets have not been identified. Actin remodeling during focal contact formation involves similar players, namely, RhoA, mDia1, and Src. Thus, although different molecules are used, the mechanisms underlying actin-dependent organelle trafficking and adhesion assembly appear to be conserved.

Myosin Ib modulates the morphology and the protein transport within multi-vesicular sorting endosomes.
Members of at least four classes of myosin (I, II, V and VI) have been implicated in the dynamics of a large variety of organelles. Despite their common motor domain structure, some of these myosins, however, are non processive and cannot move organelles along the actin tracks. Here, we demonstrate in the human pigmented MNT-1 cell line that, (1) the overexpression of one of these myosins, myos...

Microtubules are essential cytoskeletal tracks for cargo transportation in axons and also serve as the primary structural scaffold of neurons. Structural assembly, stability, and dynamics of axonal microtubules are of great interest for understanding neuronal functions and pathologies. However, microtubules are so densely packed in axons that their separations are well below the diffraction lim...
متن کاملChasing Ebola through the Endosomal Labyrinth.
During virus entry, the surface glycoprotein of Ebola virus (EBOV) undergoes a complex set of transformations within the endosomal network. Tools to study EBOV entry have been limited to static immunofluorescence or biochemical and functional analysis. In a recent article inmBio, Spence et al. reported a novel, live-cell-imaging method that tracks this transformational journey of EBOV in real t...
