Inhibition of vaccinia virus growth and virus-specific RNA synthesis by 3'-O-methyl adenosine and 3'-O-methyl guanosine.

چکیده
The 5' triphosphates of the methylated nucleoside analogs 3'-O-methyl adenosine and 3'-O-methyl guanosine are RNA chain terminators in vitro. However, anticellular or antiviral effects of 3'-O-methylated nucleosides or nucleotides have not been investigated. This is presumably because of the assumption that cellular kinases will be unable to phosphorylate the nucleosides. We report here that contrary to this assumption, 3'-O-methyl adenosine and to a lesser extent 3'-O-methyl guanosine are potent inhibitors of vaccinia virus growth in L-cells and Vero cells, without having a significant effect on cell growth at concentrations required to inhibit virus growth. Experiments revealed that early virus-specific RNA synthesis was preferentially inhibited by both 3'-O-methyl adenosine and 3'-O-methyl guanosine.

Coronavirus nonstructural protein 16 is a cap-0 binding enzyme possessing (nucleoside-2'O)-methyltransferase activity.
The coronavirus family of positive-strand RNA viruses includes important pathogens of livestock, companion animals, and humans, including the severe acute respiratory syndrome coronavirus that was responsible for a worldwide outbreak in 2003. The unusually complex coronavirus replicase/transcriptase is comprised of 15 or 16 virus-specific subunits that are autoproteolytically derived from two l...
متن کاملStructural insights into the mechanism and evolution of the vaccinia virus mRNA cap N7 methyl-transferase.
The vaccinia virus mRNA capping enzyme is a multifunctional heterodimeric protein associated with the viral polymerase that both catalyses the three steps of mRNA capping and regulates gene transcription. The structure of a subcomplex comprising the C-terminal N7-methyl-transferase (MT) domain of the large D1 subunit, the stimulatory D12 subunit and bound S-adenosyl-homocysteine (AdoHcy) has be...
متن کاملMethylated nucleotides block 5'-terminus of vaccinia virus messenger RNA.
Studies on the nature and location of the methylated nucleotides in mRNA synthesized in vitro by vaccinia virus particles revealed an unusual 5'-terminal structure. Evidence that the pyrophosphate group is blocked by 7-methylguanosine and that both 2'-O-methyl-adenosine and 2'-O-methylguanosine occupy penultimate positions was presented. According to this model, the 5'-termini of vaccinia virus...
متن کاملInhibition of Release of Vaccinia Virus by N1-isonicotinoyl-n2-3-methyl-4-chlorobenzoylhydrazine
N(1)-isonicotinoyl-N(2)-3-methyl-4-chlorobenzoylhydrazine (IMCBH) is a selective inhibitor of vaccinia virus multiplication. In concentrations up to 50 microg/ml, IMCBH causes neither toxic morphologic changes, nor does it inhibit the multiplication of cells. Viruses other than vaccinia are not affected by IMCBH. The virus-inhibitory effect of IMCBH is dependent on the type of host cell used, i...

A series of novel 2-(substitutedphenyl)-5-methyl-3-(phenylamino)-1,3-thiazolidin-4-ones (4a-g) were synthesized and structurally confirmed by elemental, IR, H NMR and MS spectral analysis. Further, the antiviral screening of 2-(substitutedphenyl)-5-methyl-3-(phenylamino)-1,3-thiazolidin-4-ones (4a-g) against a broad panel of DNA and RNA viral strains indicated that 2-(4-chlorophenyl)-5-methyl-3...
