چکیده

Photothermal excitation is an alternative cantilever drive mechanism for tapping mode atomic force microscopy. It uses a power-modulated laser focused on the cantilever to directly drive its oscillation, producing cantilever tunes that match the thermal response almost perfectly. This enables quantitatively accurate AFM imaging and viscoelastic nanomechanical mapping in all environments. In addition, the clean cantilever tunes are stable with time, even in liquids, and therefore result in exceptional imaging stability. Lastly, the ease of use of photothermal excitation and compatibility with small high-frequency cantilevers allows high-speed imaging in both air and liquid environments. We explain the advantages of photothermal excitation in these contexts, and contrast its benefits to problems associated with the well-established piezoacoustic drive

Efficiency improvement in the cantilever photothermal excitation method using a photothermal conversion layer

Photothermal excitation is a cantilever excitation method that enables stable and accurate operation for dynamic-mode AFM measurements. However, the low excitation efficiency of the method has often limited its application in practical studies. In this study, we propose a method for improving the photothermal excitation efficiency by coating cantilever backside surface near its fixed end with c...

Tapping mode atomic force microscopy in liquid

We show that standard silicon nitride cantilevers can be used for tapping mode atomic force microscopy (AFM) in air, provided that the energy of the oscillating cantilever is sufficiently high to overcome the adhesion of the water layer. The same cantilevers are successfully used for tapping mode AFhif in liquid. Acoustic modes in the liquid excite the canti1eve.r. On soft samples, e.g., biolog...

Dynamic proportional-integral-differential controller for high-speed atomic force microscopy

In tapping mode atomic force microscopy, the cantilever tip intermittently taps the sample as the tip scans over the surface. This mode is suitable for imaging fragile samples such as biological macromolecules, because vertical oscillation of the cantilever reduces lateral forces between the tip and sample. However, the tapping force vertical force is not necessarily weak enough for delicate sa...
