Reversible Metabolic Swelling of Bacterial Protoplasts* T

چکیده

In a previous communication (I) it was reported that the cell walls of Streptococcus fecalis (ATCC 9790) were susceptible to digestion by lysozyme. This finding subsequently allowed the preparation of protoplasts from these organisms by treatment of whole cells with lysozyme and the use of high concentrations of sucrose as the impermeantl substance necessary for osmotic stabilization. Further investigations led to the observation that addition of relatively small amounts of glucose to suspensions of these protoplasts produces a reversible swelling as indicated by a marked progressive fall in turbidity which eventually reverses spontaneously and returns to its initial value. It is to be noted that the reversible swelling in this case is not elicited by deliberate changes in extracellular osmotic pressure as described for BaciZZus megaterium protoplasts (2) (or other osmotically sensitive bodies), but rather by a concentration of metabolite which does not contribute significantly to the extracellular osmotic pressure, hence the term “reversible metabolic swelling.” This paper reports a study of the phenomenon of reversible swelling particularly with regard to the roles of glycolysis and specific cations.

Light-dependent osmoregulation in pea stem protoplasts. photoreceptors, tissue specificity, ion relationships, and physiological implications.

Light-induced changes in the volume of protoplasts bathed in a medium of constant osmolarity are useful indications of light-dependent cellular osmoregulation. With this in mind, we investigated the effect of light on the volume of protoplasts isolated from the elongating stems of pea (Pisum sativum) seedlings raised under red light. The protoplasts were isolated separately from epidermal peels...

Osmotic behavior of bacterial protoplasts: temperature effects.

Among protoplasts released from cells of Bacillus megaterium grown at 20, 30, or 37 C, osmotic swelling in NaCl solution at a given external osmotic pressure was greatest for protoplasts from cells grown at 20 C and least for protoplasts from cells grown at 37 C. Protoplasts from cells grown at lower temperaturs were also less stable to osmotic shock and lysed at higher external osmotic pressur...

Effects of Purified Helminthosporium maydis Race T Toxin on the Structure and Function of Corn Mitochondria and Protoplasts.

A toxin preparation from Helminthosporium maydis Race T containing several closely related molecules with apparently identical biological activities was highly active against mitochondria and protoplasts from Texas male-sterile (T) cytoplasm corn (T mitochondria and T protoplasts, respectively) but had no effect on their male-fertile (N) cytoplasm counterparts. The toxin preparation caused mult...
