چکیده

Once the cells are challenged by intraand extracellular environmental stimuli such as nitric oxide, calcium and pathogenic organisms, the oxidative balance between reactive oxygen species (ROS) (such as O 2 , H 2 O 2 , OH·) production and antioxidant defense systems (such as glutathione reductase and catalase) is broken, resulting in accumulation of ROS within cells. ROS is linked with various cellular signaling pathways, including the nuclear factor erythroid 2-related factor 2/Keap1(Nrf2/Keap1), mitogen-activated protein kinases (MAPKs), the nuclear factor kappa B (NF-κB), protein kinase C (PKC), signal transducers and activators of transcription 3 (STAT3), and peroxisome proliferator-activated receptor-γ (PPARγ), which engaged in regulating pro-oxidant genes and antioxidant genes expression, and mediate cells oxidative injury and antioxidant defense system. This article therefore mainly focuses on: 1) the production of ROS; 2) Nrf2/Keap1, MAPKs, NF-κB, PKC, STAT3, and PPARγ activation mechanism by ROS.

Advanced Glycation End-Products and Their Receptor-Mediated Roles: Inflammation and Oxidative Stress

Glycation is a protein modification, which results in a change in a protein structure. Glycation is believed to be the etiology of various age-related diseases such as diabetes mellitus and Alz-heimer’s disease (AD). Activation of microglia and resident macrophages in the brain by glycated proteins with subsequent oxidative stress and cytokine release may be an important factor in the progressi...

The Jak-Stat Signaling Pathway of Interferons System: Snapshots

Interferons (IFNs) are a family of small regulatory glycoproteins that play a central role in the defense against viral infections. Although IFNs have been initially discovered as antiviral factors, today they are known as an integral part of the cytokine network that affect a wide range of biological processes. IFNs exert their pleiotropic effects through their multisubunit cell surface recept...

Activation of Wnt signaling reduces high-glucose mediated damages on skin fibroblast cells

Objective(s): High-glucose (HG) stress, a mimic of diabetes mellitus (DM) in culture cells, alters expression of a large number of genes including Wnt and NF-κB signaling-related genes; however, the role of Wnt signaling during HG-mediated fibroblast damage and the relationship between Wnt and NF-κB signaling have not been understood. In this study, we aimed to investigate the ffects of Wnt sig...

Role of Differential Signaling Pathways and Oxidative Stress in Diabetic Cardiomyopathy

Diabetes mellitus increases the risk of heart failure independently of underlying coronary artery disease, and many believe that diabetes leads to cardiomyopathy. The underlying pathogenesis is partially understood. Several factors may contribute to the development of cardiac dysfunction in the absence of coronary artery disease in diabetes mellitus. There is growing evidence that excess genera...

The Role of AGE/RAGE Signaling in Diabetes-Mediated Vascular Calcification

AGE/RAGE signaling has been a well-studied cascade in many different disease states, particularly diabetes. Due to the complex nature of the receptor and multiple intersecting pathways, the AGE/RAGE signaling mechanism is still not well understood. The purpose of this review is to highlight key areas of AGE/RAGE mediated vascular calcification as a complication of diabetes. AGE/RAGE signaling h...

Oxidative Stress and Human Disease: a Review

Background: The human body needs both types of oxidants (free radicals) and antioxidants for normal metabolism, signal transduction mechanism, and the order of cellular activity. Free radicals can be either harmful or helpful to the body, in fact, they play a dual role as both toxic and beneficial compounds. The human body has the natural antioxidant defense mechanism to counteract the free rad...
