چکیده

A protocol for the generation of high yields of viable protoplasts has been developed for several highly branched (colonial) strains of the Quorn2 myco-protein fungus, Fusarium graminearum A3}5. Driselase was found to produce higher protoplast yields (ca 10* g−" wet weight) than the other lytic enzymes tested (Glucanex, Novozyme, β-glucuronidase, Sigma lytic enzyme, or ICN lytic enzyme), although yields differed for the various strains. Protoplast regeneration frequencies of 25–50% were observed when glucose (1±0 ) or sucrose (1±0 ) was used as the osmotic stabilizer. A highly branched strain of F. graminearum CC1-5, which grows better in submerged culture than the more sparsely branched wild-type strain (A3}5) was transformed using the hygromycin B resistance plasmid pAN7-1.

Random Insertional Mutagenesis Identifies Genes Associated with Virulence in the Wheat Scab Fungus Fusarium graminearum.

ABSTRACT Fusarium graminearum is an important pathogen of small grains and maize in many areas of the world. To better understand the molecular mechanisms of F. graminearum pathogenesis, we used the restriction enzyme-mediated integration (REMI) approach to generate random insertional mutants. Eleven pathogenicity mutants were identified by screening 6,500 hygromycin-resistant transformants. Ge...

Genetic transformation of Tomato with three pathogenesis-related protein genes for increased resistance to Fusarium oxysporum f.sp. lycopersici

Fusarium wilt caused by Fusarium oxysporum f.sp. Lycopersici is one of the major obstacles to the production of tomato which causes huge losses in tomato products worldwide. In order to increase the tolerance to this disease, a triple structure containing PR1, chitinase and glucanase genes controlled by 35S promoter was transferred to tomato. Eight days after planting on pre-culture me...

Reduced virulence of Gibberella zeae caused by disruption of a trichothecene toxin biosynthetic gene.

The production of trichothecene mycotoxins by some plant pathogenic species of Fusarium is thought to contribute to their virulence. Gibberella zeae (F. graminearum) is an important cereal pathogen that produces the trichothecene deoxynivalenol. To determine if trichothecene production contributes to the virulence of G. zeae, we generated trichothecene-deficient mutants of the fungus by gene di...

Geneticin (G418) resistance and electroporation-mediated transformation of Fusarium graminearum and F. culmorum

Fusarium graminearum and F. culmorum are phytopathogenic species causing scab and root rot diseases in all small grain cereals worldwide including Turkey. In this study, resistance levels to geneticin (G418) of 14 F. graminearum and 24 F. culmorum isolates collected from cereals were determined. Fungal cultures were grown on potato dextrose agar medium supplemented with 0, 25, 50, 75 and 100 µg...

Transformation of Potato (Solanum tuberosumcv.Savalan) by Chitinase and β-1,3-Glucanase Genes of Myco-Parasitic Fungi Towards Improving Resistance to Rhizoctonia solani AG-3

Potato (Solanum tuberosum L.) an agro-economically important food crop in the world, is sensitive to many fungal pathogens including Rhizoctonia solani (AG-3), the causal agent of stem and root rot diseases. Chitinase and glucanase are cell wall degrading enzymes which have been shown to have high antifungal activity against a wide range of phytopathogenic fungi. In the present study, plasmid p...
