چکیده

A theory of sideband intensity is derived by expanding into a Taylor series the free induction decay observed under magic angle spinning (MAS). According to this procedure, the free induction decay signal is completely represented by a basis of irreducible tensors from rank zero to rank infinity. After averaging over all orientations, only the zero-order irreducible tensors contribute to the sideband intensities. Symmetry properties of the sidebands can be seen clearly in this expansion, and an approximate formula up to ninth order is obtained by truncating the series. Sideband intensities can be calculated rapidly with this formula. The results agree satisfactorily with the exact sideband intensities obtained by numerical simulation if the ratio of the anisotropy to the spinning speed, W o6/w" is smaller than 3. The relationship of the sideband intensities with the moments of a MAS spectrum shows that the proposed method is an alternative to moment analysis when the spinning speed is not very slow. Anisotropic information about the chemical shift anisotropy interaction therefore can be extracted efficiently from experimental spectra by this approximate method.

NMR of bicelles: orientation and mosaic spread of the liquid-crystal director under sample rotation.

Model-membrane systems composed of liquid-crystalline bicellar phases can be uniaxially oriented with respect to a magnetic field, thereby facilitating structural and dynamics studies of membrane-associated proteins. Here we quantitatively characterize a method that allows the manipulation of the direction of this uniaxial orientation. Bicelles formed from DMPC/DHPC are examined by (31)P NMR un...

Identifying anisotropic constraints in multiply labeled bacteriorhodopsin by 15N MAOSS NMR: a general approach to structural studies of membrane proteins.

Structural models of membrane proteins can be refined with sets of multiple orientation constraints derived from structural NMR studies of specifically labeled amino acids. The magic angle oriented sample spinning (MAOSS) NMR approach was used to determine a set of orientational constraints in bacteriorhodopsin (bR) in the purple membrane (PM). This method combines the benefits of magic angle s...

High-resolution (1)H NMR spectroscopy in organs and tissues using slow magic angle spinning.

It is shown that high-resolution (1)H NMR spectra of intact excised tissues and organs can be obtained by rotating the sample slowly about an axis at the magic angle of 54 degrees 44' with the external magnetic field. In this way tissue and cellular damage invoked by standard magic angle spinning (MAS) experiments, where spinning speeds of several kHz are typically employed, are minimized. Spec...

Magnetization transfer magic-angle-spinning z-spectroscopy of excised tissues.

NMR experiments devised to aid in analyses of tissues include magnetization transfer (MT), which can highlight the signals of biological macromolecules through cross-relaxation and/or chemical exchange processes with the bulk (1)H water resonance, and high-resolution magic-angle-spinning (HRMAS) methods, akin to those used in solid-state NMR to introduce additional spectral resolution via the a...

Slow-spinning low-sideband HR-MAS NMR spectroscopy: delicate analysis of biological samples

High-Resolution Magic-Angle Spinning (HR-MAS) NMR spectroscopy has become an extremely versatile analytical tool to study heterogeneous systems endowed with liquid-like dynamics. Spinning frequencies of several kHz are however required to obtain NMR spectra, devoid of spinning sidebands, with a resolution approaching that of purely isotropic liquid samples. An important limitation of the method...
