Application of the phenol-hypochlorite reaction to measurement of ammonia concentrations in Kjeldahl digests of serum and various tissues.

چکیده
Conditions have been defined that enable the use of nitroprusside as a catalyst for the phenol-hypochiorite reaction when the latter is applied to measurement of nitrogen in Kjeldahl digests containing mercury. This is accomplished by lowering the concentration of mercury to a range where it remains effective in accelerating the destruction of organic matter during digestion but no longer reacts with nitroprusside. In addition, the use of minimal concentrations of nitroprusside diminishes further the possibility of interference by mercury and has the added advantage of decreasing the pH dependence of indophenol blue formation. The sensitivity of the reaction makes possible measurement of nitrogen in very small samples with relatively short digestion times.

Coulometric Determination of Protein Nitrogen. Application to Direct Titration of Kjeldahl Digests.
A coulometric method is described for the determination of ammonia resulting from micro-Kjeldahl digestion of protein samples. The method has been applied to the direct titration of the ammonia in the digests. Routine titration of the ammonia resulting from lOsl. serum samples is described; the method has been applied to 1 -JLI. samples. Results are compared with standard macroand micro-Kjeldah...
متن کاملDetermination of Ammonia in Blood by Donald D. Van Slyke and Alma Hiller with the Technical Amstance of John Plazin
In the method here presented Nash and Benedict’s (1921) procedure for aerating the ammonia from blood into an acid-receiving solution is utilized without modification. For determining the ammonia obtained, however, we have employed, instead of Nesslerization, the blue color developed when ammonia is heated with an alkaline solution of phenol and hypochlorite. This reaction was apparently first ...
متن کاملProgress toward the development of a point-of-care photonic crystal ammonia sensor.
We have developed an ammonia-sensitive material by coupling the Berthelot reaction to our polymerized crystalline colloidal array (PCCA) technology. The material consists of a periodic array of highly charged colloidal particles (110 nm diameter) embedded in a poly(hydroxyethyl acrylate) hydrogel. The particles have a lattice spacing such that they Bragg-diffract visible light. In the Berthelot...
متن کاملColorimetric determination of serum acid phosphatase activity using adenosine 3'-monophosphate as substrate.
The hydrolysis of adenosine 3'-monophosphate by serum acid phosphatase has been coupled to the liberation of ammonia from the adenosine generated through the action of exogenous adenosine deaminase. The ammonia is measured at the end of the incubation by a modification of the phenol-hypochlorite reaction of Berthelot. Optimum conditions for the enzyme reaction have been defined. Inhibition of t...
متن کاملAn automated microassay for blood ammonia.
A microassay for blood ammonia has been developed. After dilution, blood is dialyzed by a continuous flow procedure, and the ammonium ions in the dialysate are assayed colorimetrically by the hypochlorite-phenol Berthelot reaction. Concentrations from 1 to 400 mumol/l NH+4 can be measured in a 50 mul whole blood sample. Using slight modifications, concentrations up to 800 mumol/l NH+4 can be de...
