چکیده
Ole Andreas Andersen received hisM.Sc and Ph.D from theUniversity of Tromsø, Norway, in 1998 and 2003, respectively. His Ph.Dwork was supervised by Prof. Edward Hough and comprised crystallographic and biochemical studies on human phenylalanine hydroxylase. Andersen had a postdoctoral fellowship for one year at The Norwegian Structural Biology Centre in Tromsø under the supervision of Prof. Arne Smalås. He is currently working on chitinases using protein crystallography as a tool for inhibitor design in the group of Daan van Aalten at the University of Dundee.

Structure-Based Dissection of the Natural Product Cyclopentapeptide Chitinase Inhibitor Argifin
Chitinase inhibitors have chemotherapeutic potential as fungicides, pesticides, and antiasthmatics. Argifin, a natural product cyclopentapeptide, competitively inhibits family 18 chitinases in the nanomolar to micromolar range and shows extensive substrate mimicry. In an attempt to map the active fragments of this large natural product, the cyclopentapeptide was progressively dissected down to ...
متن کاملHigh-resolution structures of a chitinase complexed with natural product cyclopentapeptide inhibitors: mimicry of carbohydrate substrate.
Over the past years, family 18 chitinases have been validated as potential targets for the design of drugs against human pathogens that contain or interact with chitin during their normal life cycles. Thus far, only one potent chitinase inhibitor has been described in detail, the pseudotrisaccharide allosamidin. Recently, however, two potent natural-product cyclopentapeptide chitinase inhibitor...
متن کاملNatural product-guided discovery of a fungal chitinase inhibitor.
Natural products are often large, synthetically intractable molecules, yet frequently offer surprising inroads into previously unexplored chemical space for enzyme inhibitors. Argifin is a cyclic pentapeptide that was originally isolated as a fungal natural product. It competitively inhibits family 18 chitinases by mimicking the chitooligosaccharide substrate of these enzymes. Interestingly, ar...
متن کاملSpecificity and affinity of natural product cyclopentapeptide inhibitors against A. fumigatus, human, and bacterial chitinases.
Family 18 chitinases play key roles in organisms ranging from bacteria to man. There is a need for specific, potent inhibitors to probe the function of these chitinases in different organisms. Such molecules could also provide leads for the development of chemotherapeuticals with fungicidal, insecticidal, or anti-inflammatory potential. Recently, two natural product peptides, argifin and argadi...
متن کاملScreening-based Discovery and Structural Dissection
Family 18 chitinases play key roles in the life cycles of a variety of organisms ranging from bacteria to man. Very recently it has been shown that oneof themammalian chitinases is highly overexpressed in the asthmatic lung and contributes to the pathogenic process through recruitment of inflammatory cells. Although several potent natural product chitinase inhibitors have been identified, their...
متن کاملAn efficient synthesis of argifin: a natural product chitinase inhibitor with chemotherapeutic potential.
The first synthesis of the cyclopentapeptide family 18 chitinase inhibitor argifin has been achieved by a combination of solid phase and solution chemistry. Synthetic argifin is a nanomolar inhibitor of chitinase B1 from Aspergillus fumigatus and the high-resolution X-ray structure of the synthesized material in complex with the same enzyme is identical to that previously obtained for the natur...
