Use of organic solvents in cylinder-plate assays.

چکیده
A new antibiotic, aterrimin (Lewis et al., 1953), is unstable in aqueous solution at pH 7, and insoluble at lower pH levels. Aterrimin can be dissolved in absolute alcohol and the solutions can be used satisfactorily in a cylinder-plate type of assay (Heatley, 1944). Further investigation has established that sevbral other organic solvents are nontoxic to various bacteria when used in cylinders. This information may prove useful to others who wish to assay antibiotics which are not readily soluble or which are unstable in aqueous solutions. The literature suggests that it has been generally assumed that high concentrations of organic solvents would interfere with cylinder-plate assays. No referlarly of crude preparations which contain viable spores from the parent culture, the paper-disc method affords less protection against contamination than the cylinderplate method. The purification of aterrimin has been followed by cylinder-plate assays with Bacillus megaterium NRRL B-938 on nutrient agar (Difco) plates incubated for 16 hours at 35 C. Samples were evaporated when necessary and dissolved in absolute ethyl alcohol; or they were diluted with ethyl alcohol so that the concentration of any other solvent originally present was reduced to less than 3 per cent. Standards were prepared in absolute ethyl alcohol. Inconsistent results may arise if all samples and standards are not added as solutions

Counting Non-Aqueous Samples in the TopCount® Microplate Scintillation and Luminescence
The microplate format has become a standard for many biological assays such as high throughput screening for drug discovery and radioimmunoassays. Productivity with these high volume procedures has been limited by the amount of sample handling required and low sample counting throughput. Now, however, microplate assays can be efficiently counted in microplates with the Packard TopCount Micropla...
متن کاملUSE OF ORGANIC SOLVENTS FOR THE ENHANCEMENT AND LIFETIME INCREASING OF LUMINESCENCE IN FLUORIMETRIC APPLICATION BY FLUORESCAMINE
In this study, the fluorogenic efficiency of fluorescamine for spectrofluorimetric analysis of primary amines was investigated. Different mixtures of dichloromethane and other organic solvents were used for evaluation of fluorescence stability and intensity of fluorescamine derivative (Pyrrolinone). Solvents such as acetonitrile (ACN), tetrahydrofuran (THF) and dioxane increase stability an...
متن کاملAn Experimental Study of Compound Control Method around a Cylinder
In this work, the compound flow control method (passive and active) has been described. EHD actuators as wire-plate (active) and splitter plate (passive) were coupled and applied to control fluid flow and heat transfer around cylinder in cross flow. Investigation consists of the interaction between electric field, fluid flow and temperature field. Experimental tests included various positions o...
متن کاملPredicting Solubility of Isotactic Poly (1-Butene) in Different Organic Solvents by Modified UNIQUAC
The UNIQUAC activity coefficient model is extended to predict solid-liquid equilibrium of isotactic crystalline poly (1-butene), iPBu-1, in different organic solvents. The UNIQUAC activity coefficient model used in this work is based on the concept of group contribution and consists of combinatorial, residual and free volume terms. To account for branching of atom groups in the monomer structur...

Introduction: Simultaneous development of technology is composed with environmental hazardous with heavy-metals and organic solvents and thus toxic catalysts/organic solvents must be replaced with biocatalysts and water. Urease is a hydrolase enzyme and urease-urea can be considered a safe source of toxic ammonia in synthesis of nifedipine. Nifedipine is an antihypertensive drug synthesized by ...
