Properties of highly purified fucan.

چکیده
Fucan was first isolated by Kylin from brown seaweed and named fucoidin by him (1). More recently fucan was isolated by Percival and Ross (2) from several Phaeophyceae. They considered it to be a polymeric salt of fucose monosulfate. However, their most highly purified preparation still contained approximately 5.6% sugars other than L-fucose and 3.3 ‘% uranic acid. A branched structure was suggested (3) for fucan with 1,2 links predominating (3, 4). The molecular weight of fucan determined by osmometric means was 133,000 f 20,000 (4). Anticoagulant active fractions were isolated from crude fucan in a yield of about 25% (5). Other fractions, although less active as anticoagulants, were potent lipemia clearers (6). All of these fractions contained a still unidentified sugar with chromatographic properties similar to but not identical with 3-O-methylfucose (5, 7). In the present work, highly purified fractions were prepared from crude fucan and their physical, chemical, and biological properties investigated. It is hoped that the study of physical behavior and the chemistry of these fucan fractions will contribute to a better understanding of the relation of biological activity to physical and chemical characteristics of fucan and other sulfated high polymers.

Females of the sea urchin Strongylocentrotus purpuratus differ in the structures of their egg jelly sulfated fucans.
The egg jelly coats of sea urchins contain sulfated fucans which bind to a sperm surface receptor glycoprotein to initiate the signal transduction events resulting in the sperm acrosome reaction. The acrosome reaction is an ion channel regulated exocytosis which is an obligatory event for sperm binding to, and fusion with, the egg. Approximately 90% of individual females of the sea urchin Stron...

Background and Objectives: Laccase is the most abundant member of protein family that catalyzes the oxidation of substituted phenols. Laccases are used as biocatalysts for decolorization and bleaching in dye industries, detoxification in environment, and juice clarification in food industries. The present study aimed at producing recombinant laccase, purifying with high yield and fold, and char...
متن کاملMonoclonal Antibodies Directed to Fucoidan Preparations from Brown Algae
Cell walls of the brown algae contain a diverse range of polysaccharides with useful bioactivities. The precise structures of the sulfated fucan/fucoidan group of polysaccharides and their roles in generating cell wall architectures and cell properties are not known in detail. Four rat monoclonal antibodies, BAM1 to BAM4, directed to sulfated fucan preparations, have been generated and used to ...
متن کاملEffects of polysaccharides enriched in 2,4-disulfated fucose units on coagulation, thrombosis and bleeding. Practical and conceptual implications.
Sulfated polysaccharides from marine invertebrates have well-defined structures and constitute a reliable class of molecules for structure-activity relationship studies. We tested the effects of two of these polysaccharides, namely a sulfated fucan and a fucosylated chondroitin sulfate, on coagulation, thrombosis and bleeding. The compounds share similar 2,4-disulfated fucose units, which are r...
متن کاملA novel antivirally active fucan sulfate derived from an edible brown alga, Sargassum horneri.
A novel fucan sulfate (Hor-1) was isolated from the hot water extract of an edible brown alga, Sargassum horneri (Turner) C. Agardh. The fucan sulfate was revealed to have sugar linkage types, sulfate content and uronic acid content different from those of sodium hornan (Na-HOR), another fucan sulfate isolated from this alga. However, it exhibited inhibitory activity against replication of herp...
