چکیده

Ubiquitin-lysozyme conjugates have been used as substrates to identify an ATP-dependent protease from rabbit reticulocyte lysates. The enzyme, which has been partially purified by DEAE chromatography and glycerol gradient centrifugation, has an apparent molecular weight greater than 600,000 based on sedimentation and gel filtration. Whereas it degrades conjugated lysozyme molecules in the presence of ATP, the protease does not degrade free lysozyme molecules even upon addition of ubiquitin, lysozyme-ubiquitin conjugates, and ATP. Degradation of lysozyme conjugates is independent of added ubiquitin and occurs in fractions incapable of ubiquitin conjugation. Proteolysis is maximal at pH 7.8, inhibited by hemin, N-ethylmaleimide, or aurintricarboxylic acid, and proceeds with an apparent Arrhenius activation energy in the range of 27 +/- 5 kcal/mol. These properties are similar to those observed for the degradation of lysozyme conjugates in lysates indicating that the partially purified protease catalyzes the "second" ATP-utilizing reaction identified previously (Hough, R., and Rechsteiner, M. (1984) Proc. Natl. Acad. Sci. U. S. A. 81, 90-94; Hershko, A., Leshinsky, E., Ganoth, D., and Heller, H. (1984) Proc. Natl. Acad. Sci. U. S. A. 81, 1619-1623; Tanaka, K., Waxman, L., and Goldberg, A. L. (1983) J. Cell Biol. 96, 1580-1585).

ATP stimulates proteolysis in reticulocyte extracts by repressing an endogenous protease inhibitor.

An endogenous inhibitor of the reticulocyte ATP-dependent proteolytic system has been purified partially by ammonium sulfate precipitation from rabbit reticulocyte and erythrocyte extracts. Inhibitor-free protease rapidly degrades 21-40% of the substrate [14C]methyl-alpha-casein per hour, resembling ATP-dependent activity in reticulocyte extracts. This proteolytic activity is not stimulated by ...

Ubiquitin is the ATP-dependent proteolysis factor I of rabbit reticulocytes.

A small heat-stable polypeptide, ATP-dependent proteolysis factor 1 (APF-1), is an essential component of the ATP-dependent proteolytic system of rabbit reticulocytes (Ciechanover, A., Hod, Y., and Hershko. A. (1978) Biochem. Biophys. Res Commun. 81, 1100-1105). The following evidence supports the view that APF-1 is ubiquitin, a highly conserved heat-stable polypeptide found universally in natu...

Characterization of the Heat - stable Polypeptide of the ATP - dependent Proteolytic System

The energy dependence of intracellular protein breakdown has been recognized for a long time (1,2), but its biochemical mechanisms have begun to be elucidated only recently. This has been initiated by the establishment of several cell-free systems in which protein breakdown is stimulated by ATP (37 ) . We have found previously that the ATP-dependent proteolytic system from reticulocytes is comp...

Skeletal muscle and liver contain a soluble ATP + ubiquitin-dependent proteolytic system.

Although protein breakdown in most cells seems to require metabolic energy, it has only been possible to establish a soluble ATP-dependent proteolytic system in extracts of reticulocytes and erythroleukemia cells. We have now succeeded in demonstrating in soluble extracts and more purified preparations from rabbit skeletal muscle a 12-fold stimulation by ATP of breakdown of endogenous proteins ...

Some properties of 2-5A binding/nucleolytic activities in gel filtered rabbit reticulocyte lysates.

Rabbit reticulocyte lysates, gel filtered on Sephadex G-25 with or without ATP (or its analogs), were preincubated at 37 degrees C and their subsequent binding to p3A4,3'-[32P]pCp was studied. Lysates filtered without ATP or in the presence of 0.1 mM 8-bromo-ATP, 1,N6-etheno-ATP, or ITP showed a time-dependent decrease in binding activity. This decrease was completely prevented when lysates wer...
