Evaluation of Apoptotic and Growth Inhibitory Activity of Phloretin in BGC823 Gastric Cancer Cell

چکیده
Purpose: To evaluate the in vitro anti-proliferative activity and probable mechanism of phloretin in human gastric cancer BGC823 cell lines. Methods: Phloretin was isolated from apple tree leaves and identified by H-Nuclear Magnetic Resonance (NMR), C-NMR and electrospray ionization tandem mass spectrometry (ESI-MS). The inhibitory activity of the compound in BGC823 gastric cancer cells was determined by Methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay and soft agar colony formation assay while its apoptotic activity was detected by fluorescene staining and Western blotting analysis. Results: The results show that the inhibitory activity of phloretin in BGC823 gastric cancer cells was mediated by induction of apoptosis and down-regulation of Bcl2 (B-cell lymphoma 2) expression. The anti-proliferative effects of phloretin was dose-dependent and inhibited the growth of BGC823 gastric cancer cells by 73 % at 30 μM; this effectively induced cleavage of anti-poly (ADP-ribose) polymerase (PARP) as well as downregulation of Bcl2 protein expression in BGC823 cells after 24 h treatment. Conclusion: Phloretin is a promising preventive and therapeutic agent for gastric cancer.

Evaluation of Growth Inhibitory and Apoptosis Inducing Activity of Human Calprotectin on the Human Gastric Cell Line (AGS)
Background: Calprotectin is cytotoxic agent that its anti-tumor effects are governed through suppression of topoisomerase II a key enzyme in apoptosis. In previous studies, cytotoxicity and apoptotic effects of calprotectin are shown on different cancer cell lines, but not human gastric cancer cell lines. In the present study, cytotoxicity and apoptotic effects of calprotectin on human gastric ...

Background: Conventional anti-cancer treatments, such as surgery, chemotherapy, radiation therapy, etc., are linked to antimicrobial resistance (AMR) and have negative effects on healthy tissues. Objectives: The flow cytometry technique was used in this study to evaluate the effectiveness of a modified anti-cancer peptide in treating the AGS gastric cancer cell line. Methods: This study used ...

متن کاملCecropinXJ inhibits the proliferation of human gastric cancer BGC823 cells and induces cell death in vitro and in vivo.
We have shown that an antimicrobial peptide (AMP) cecropinXJ isolated from the larvae of Bombyx mori selectively inhibits the proliferation of cancer cells. However, the mechanism remains to be determined. In the present study, we examined the antitumor activity of cecropinXJ against human gastric cancer BGC823 cells and explored the mechanism. The results showed that cecropinXJ inhibited the g...

Background: Lactoferrin is a glycoprotein with a molecular weight of 80 kDa, as a member of the transferring family. In recent investigations the important biological properties such as anti microbial, antiviral and anticancer activity of lactoferrin were studied. In the present study, the effect of antitumor induced by lactoferin was evaluated on stomach cancer cell AGS. Materials and Methods...
