چکیده
At 0”, native tetrameric rabbit muscle glyceraldehyde 3-phosphate dehydrogenase (7.4 S) undergoes a reversible inactivation and dissociation into 4.4 S dimers or 3.2 S monomers, depending upon the species of salt present and upon the protein concentration. These dissociation processes have been studied using sucrose density gradient centrifugation. With 0.10 mg per ml of enzyme in 0.15 M ammonium sulfate, 4.4 S dimers predominate; with 0.05 mg per ml of enzyme in 0.15 M KCl, 3.2 S monomers predominate. At other protein concentrations in the two systems, mixtures are produced. KC1 gives greater dissociation at all protein concentrations. It also destabilizes at high salt concentrations (1.0 to 2.5 M). In contrast, destabilization in ammonium sulfate is maximal at 0.15 M; 0.5 and 1.0 M ammonium sulfate stabilize against dissociation essentially completely. The native enzyme in the absence of salts shows very slow, very slight dissociation into dhners. Inactivation and dissociation do not occur at 23’ in 0.15 M ammonium sulfate and only slightly in 0.15 M KCI, and then only at very low protein concentrations. Inactivation and dissociation are reversed by warming to 23’ and greater than 80% of the activity is recovered. The reversible dissociation and inactivation occur at both pH 7.0 (0.1 M imidazole) and at pH 9.5 (0.2 M glycine). A reducing agent (0.1 M ,&mercaptoethanol) has to be present for activity to be recovered. These data prove that this enzyme at 0’ exists as an equilibrium mixture which favors association to tetramers at high protein concentrations and dissociation into dimers or monomers or both at low protein concentrations. The dimers and monomers appear to be fairly compact.

A study of some of the physical properties of glyceraldehyde-3-phosphate dehydrogenase.
Preparation of Enzyme SolutionsRabbit muscle glyceraldehyde-3-phosphate dehydrogenase was prepared as described previously (1) and stored at 0” as the crystalline suspension. For use, the crystals were separated by centrifugation and dissolved in buffer. The resulting solution was dialyzed overnight against a flowing stream of buffer in order to remove the ammonium sulfate, equilibrated against...
متن کاملThe hybridization of glyceraldehyde 3-phosphate dehydrogenases from rabbit muscle and yeast. Kinetics and thermodynamics of the reaction and isolation of the hybrid.
A kinetic and thermodynamic study was made of the formation of the hybrid (R(2)Y(2)) glyceraldehyde 3-phosphate dehydrogenase from the yeast (Y(4)) and rabbit (R(4)) enzymes. The values of the thermodynamic parameters for the equilibrium between R(4), Y(4) and R(2)Y(2) suggest that the R(2)-R(2) and Y(2)-Y(2) interactions are similar. However, the failure to observe the RY(3) and R(3)Y hybrids ...
متن کاملThe glyceraldehyde 3-phosphate dehydrogenases of liver and muscle. Cooperative interactions and conditions for functional reversibility.
A method is described for the isolation of glyceraldehyde 3.phosphate dehydrogenase from rabbit liver. The enzyme has been crystallized as the NAD complex and its chemical and physical properties have been compared with those of the muscle enzyme. The kinetics of the reversible reaction catalyzed by the dehydrogenases is sensitive to temperature and ionic strength and has been examined at 37, p...
متن کاملFolding and association of triose phosphate isomerase from rabbit muscle.
The enzymatic activity and quaternary structure of rabbit muscle triose phosphate isomerase remains unchanged in the concentration range from 2 micrograms/ml to 2 ng/ml. In this concentration range the enzyme can be reactivated after dissociation and denaturation in 6.5 M guanidine hydrochloride. Removal of the denaturant by dilution and separation of inactive wrong aggregates (5-20%) lead back...
متن کاملDesigning transthyretin mutants affecting tetrameric structure: implications in amyloidogenicity.
The molecular mechanisms that convert soluble transthyretin (TTR) tetramers into insoluble amyloid fibrils are still unknown; dissociation of the TTR tetramer is a pre-requisite for amyloid formation in vitro and involvement of monomers and/or dimers in fibril formation has been suggested by structural studies. We have designed four mutated proteins with the purpose of stabilizing [Ser(117)-->C...
