Brief Communication Characterization of /3j- and /32-Adrenoceptor Subtypes in the Rat Atrioventricular Node by Quantitative Autoradiography

چکیده
We characterized /3-adrenoceptor subtypes in the atrioventricular node of the rat heart by quantitative autoradiography. Consecutive 16-/xm-thick sections from single rat hearts containing the atrioventricular node were incubated with increasing concentrations of [I]iodocyanopindolol. After exposure to [H]Ultrofilm, optical densities corresponding to the atrioventricular node were determined by computerized densitometry after comparison with [I]standards. The computer program LIGAND was used for analysis of receptor subtypes. Delineation of /3-adrenoceptor subtypes was achieved by incubating consecutive tissue sections with 50 pM [I]iodocyanopindolol in the presence of increasing concentrations of the /3,-selective antagonist atenolol or the /3,-selective antagonist ICI 118,551. The atrioventricular node contains a higher concentration of /3-adrenoceptors than the adjacent interventricular septum. We estimated that the proportions of /3,and /3,-adrenoceptors in the atrioventricular node were about 56% and 44% of the total binding capacity respectively. (Circulation Research 1988;62:173-177)

متن کاملCharacterization of angiotensin II receptor subtypes in rat heart.
Angiotensin II exerts positive inotropic and chronotropic effects on the mammalian heart by binding to specific membrane receptors. Recently, two subtypes of angiotensin II receptors (AT1 and AT2) have been distinguished by using the nonpeptide antagonists losartan (previously known as DuP 753) and PD123177. To evaluate the tissue distribution and subtypes of angiotensin II receptors in rat hea...
متن کاملHistological study of the atrioventricular node and bundle in the heart of ovine fetus
This study was conducted on the atrioventricular node (AVN) and atrioventricular bundle (AVB) of fivefour-month-old ovine fetuses. The histological structure of these components was studied by routinehistological techniques and use of specific staining methods. The AVN was caudally located adjacent to theroot of the aorta. It was almost spherical in shape and consisted of twisty cells. The node...
متن کاملThe role of Na+-K+-ATPase in the basic and rate-dependent properties of isolated perfused rabbit Atrioventricular Node
Introduction: Ouabaine is a well-known atrioventricular (AV) node depressant agent, but its effects on functional properties of the AV node have not been cleared. The aim of the present study was to determine how ouabaine administration modifies the rate-dependent properties of the AV node. Methods: Selective stimulation protocols were used to quantify independently electrophysiological prop...
متن کاملModulation of extracellular atrioventricular node field potential pattern and ventricular rhythm by morphine in experimental atrial fibrillation in isolated rabbit heart
Introduction: Endorphins are produced by cardiomyocytes, and exert different effects on the heart. The aim of the present study is to assess morphine effects on extracellular atrioventricular (AV) node field potential pattern and ventricular rhythm of isolated rabbit heart during experimental atrial fibrillation (AF). Methods: Effects of different concentrations of morphine (10, 20, 50 and 1...
