Inhibition of choline acetyltransferase as a mechanism for cholinergic dysfunction induced by amyloid-β oligomers

چکیده
Background: Cholinergic dysfunction is an early feature of Alzheimer’s disease (AD). Results: Soluble oligomers of the amyloid-β peptide (Aβ) bind to cholinergic neurons and inhibit choline acetyl transferase (ChAT) activity, before any cell death or lesion. Conclusion: ChAT inhibiton might impair acetylcholine production and cholinergic function in AD brains. Significance: This novel effect of Aβ oligomers may be relevant in early stage AD pathology.

Inhibition of choline acetyltransferase as a mechanism for cholinergic dysfunction induced by amyloid-β peptide oligomers.
Dysregulated cholinergic signaling is an early hallmark of Alzheimer disease (AD), usually ascribed to degeneration of cholinergic neurons induced by the amyloid-β peptide (Aβ). It is now generally accepted that neuronal dysfunction and memory deficits in the early stages of AD are caused by the neuronal impact of soluble Aβ oligomers (AβOs). AβOs build up in AD brain and specifically attach to...

The effect of β-amyloid aggregates on activity of choline acetyltransferase (ChAT) which is responsible for synthesizing acetylcholine (ACh) in human brain is investigated through the two-enzyme/two-compartment (2E2C) model where the presynaptic neuron is considered as compartment 1 while both the synaptic cleft and the postsynaptic neuron are considered as compartment 2 through suggesting thre...
متن کاملThe basal forebrain cholinergic system in aging and dementia. Rescuing cholinergic neurons from neurotoxic amyloid-β42 with memantine.
The dysfunction and loss of basal forebrain cholinergic neurons and their cortical projections are among the earliest pathological events in the pathogenesis of Alzheimer's disease (AD). The evidence pointing to cholinergic impairments come from studies that report a decline in the activity of choline acetyltransferase (ChAT) and acetylcholine esterase (AChE), acetylcholine (ACh) release and th...
متن کاملEffect of nicotine on neuronal dysfunction induced by intracerebroventricular infusion of amyloid-β peptide in rats.
OBJECTIVE The aim of the study was to investigate the effects of nicotine on learning and memory deficits induced by intracerebroventricular infusion of amyloid-β peptide (Aβ) in rats. MATERIALS AND METHODS Neuronal dysfunction in rats was induced by an infusion of Aβ(1-42) (20 µg/body, over 3 days) into right ventricle. Nicotine was administered intraperitoneally to the rats at 0.2 mg/kg, on...
متن کاملNeuroprotection of N-benzylcinnamide on scopolamine-induced cholinergic dysfunction in human SH-SY5Y neuroblastoma cells
Alzheimer's disease, a progressive neurodegenerative disease, affects learning and memory resulting from cholinergic dysfunction. Scopolamine has been employed to induce Alzheimer's disease-like pathology in vivo and in vitro through alteration of cholinergic system. N-benzylcinnamide (PT-3), purified from Piper submultinerve, has been shown to exhibit neuroprotective properties against amyloid...
