چکیده
Microbial degradation of cellulose and lignocelluloses derivatives has received considerable attention towards bioremediation in recent years. The biological significance of lignin combined with the commercial utility of lignocellulose has generated widespread interest in understanding the biochemistry of lignocellulose degradation which is carried out by one of the copper containing oxidative enzyme namely Laccase. These enzymes are present in all organisms which are involved in lignocellulose degradation but their copper binding sites in the respective amino acid residues are still controversial. In order to overcome this puzzle: sequence analysis was performed in selected lignocellulose degrading fungus namely Aspergillus fumigatus, Aspergillus flavus and Fusarium oxysporum to understand the evolutionary relationship via Multalign, Phylogeny.fr and MetalDetector. This study reveals the evolutionary relationship of Laccase in the above fungal sps., involved in lignocellulose degradation clearly shows: evolution pattern of Laccase in selected fungus with high bootstrap value. A.Flavus Laccase which holds the molecular weight of 65706.2 Da is the ancestor for subsequent evolution of Laccase in other fungal sps and it Metal binding residues identification to enhance the Laccase activity in selected microorganisms against organic waste contaminant area and to increase the process of lignocellulose degradation towards biofuel, composting and bioremediation or Solid waste management applications.

Diversity of two-domain laccase-like multicopper oxidase genes in Streptomyces spp.: identification of genes potentially involved in extracellular activities and lignocellulose degradation during composting of agricultural waste.
Traditional three-domain fungal and bacterial laccases have been extensively studied for their significance in various biotechnological applications. Growing molecular evidence points to a wide occurrence of more recently recognized two-domain laccase-like multicopper oxidase (LMCO) genes in Streptomyces spp. However, the current knowledge about their ecological role and distribution in natural...
متن کاملCharacterization, molecular cloning, and differential expression analysis of laccase genes from the edible mushroom Lentinula edodes.
The effect of different substrates and various developmental stages (mycelium growth, primordium appearance, and fruiting-body formation) on laccase production in the edible mushroom Lentinula edodes was studied. The cap of the mature mushroom showed the highest laccase activity, and laccase activity was not stimulated by some well-known laccase inducers or sawdust. For our molecular studies, t...
متن کاملIdentification of metallothionein gene structure in sterlet (Acipenser ruthenus)
Aquatic organisms present, not only simple sources of accumulated metal, but can interact with metals, altering their toxicity. Due to exposition of biosphere with metals, organisms have developed various defense mechanisms to protect themselves against adverse effects of these ions and their compounds. Metallothionein (MT) is one of that which represents a critical mechanism for detoxification...
متن کاملSymbiotic fungi produce laccases potentially involved in phenol degradation in fungus combs of fungus-growing termites in Thailand.
Fungus-growing termites efficiently decompose plant litter through their symbiotic relationship with basidiomycete fungi of the genus Termitomyces. Here, we investigated phenol-oxidizing enzymes in symbiotic fungi and fungus combs (a substrate used to cultivate symbiotic fungi) from termites belonging to the genera Macrotermes, Odontotermes, and Microtermes in Thailand, because these enzymes ar...
