Structural aspects of ribonucleoprotein interactions in ribosomes

چکیده
The increasing sophistication in instrumentation, the implementation of powerful genetic techniques and the use of ribosomes of halophilic and thermophilic bacteria have resulted in major accomplishments which could not be foreseen earlier. Most of the recent studies are reviewed in three volumes of Biochemie, two of which are dedicated to the late HG Wittmann and one to A Spirin' s retirement, as well as in the proceedings of the International Conference on 'The Translational Appara ms' held in Berlin in November 1992, which below is called 'Ribosome-92'. We have selected a subset of studies which, in our opinion, reflects the characteristic trends in the past year's structural research in protein-RNA interactions. Studies on ribosomes as a cellular assemblage will be addressed in the next issue of Current Opinion in Structural Biology. face of these subunits (E Arndt, C Paulke, F Franceschi, abstract 86, Ribosome-92); and the crystallization of particles lacking a few proteins [4"].

Ribosome processing in HeLa cells. Studies on structural aspects of precursor and mature ribosomes
Mature ribosomes, their subunits, and their ribonucleoprotein precursors were extracted from HeLa cells and prepared for electron microscopy by a method based on adsorption of nucleic acid to a charged grid. Images so obtained revealed the presence of two types of ribonuclease-sensitive fibrils of consistent morphology. One of these types of fibril is present on the 80S and 55S nucleolar ribonu...

1. After incorporation of [(14)C]valine in vitro, cerebral microsomes were separated into membrane-bound and free ribosomes by sucrose-density-gradient centrifugation. 2. In preparations from both 4-day-old and adult rats, free and bound ribosomes incorporated [(14)C]valine. Free ribosomes could be found as polysomes, which were highly active. 3. Microsomes labelled with [(14)C]valine in vitro ...

Protein synthesis is one of the most complex cellular processes, involving numerous translation components that interact in multiple sequential steps. The most complex stage in protein synthesis is the initiation process. The basal set of factors required for translation initiation has been determined, and biochemical, genetic, and structural studies are now beginning to reveal details of their...
متن کاملEffect of Proteolytic Enzymes on Sedimentation Properties of Ribonucleoprotein Particles from Heart Muscle.
There is much support for the view that the functional unit of protein synthesis is an aggregate of ribosomes held together by a strand of messenger RNA.1-6 The concept derives from the observation that radioactive amino acids are preferentially incorporated into ribosomal aggregates (termed polyribosomes, polysomes, or ergosomes) and that treatment of the ribosomal aggregates with small amount...
متن کاملAssembly factors Rpf2 and Rrs1 recruit 5S rRNA and ribosomal proteins rpL5 and rpL11 into nascent ribosomes.
More than 170 proteins are necessary for assembly of ribosomes in eukaryotes. However, cofactors that function with each of these proteins, substrates on which they act, and the precise functions of assembly factors--e.g., recruiting other molecules into preribosomes or triggering structural rearrangements of pre-rRNPs--remain mostly unknown. Here we investigated the recruitment of two ribosoma...
