Improved manual spectrophotometric procedure for determination of serum triglycerides.

چکیده
We have modified the procedure of Levy [Ann. C/in. Lab. Sd. 6, 474 (1972)] for determination of serum triglycerides. The changes consist of using heptane in place of normane; using concentrated aqueous potassium hydroxide instead of sodium methylate as the saponifying reagent; and a somewhat higher saponification, oxidation, and condensation temperature. Consequently, the yellow dehydrolutidine derivative is determined in a homogeneous solution having an almost linear absorbance up to 3 g/liter.

Modification of the DuPont aca as exemplified by a new procedure for neonatal total bilirubin.
Hemoglobin interferes with determination of total bilirubin by the diazo procedure with the DuPont aca. We compared results by that procedure with those by a manual direct spectrophotometric two-wavelength method. Because the manual method was not inhibited by hemolysis as was the DuPont aca diazo procedure, we reprogrammed our DuPont aca to measure the absorbance of diluted serum samples at 45...
متن کاملA NEW SPECTROPHOTOMETRIC PROCEDURE FOR THE DETERMINATION OF NICKEL (11) USING 6-PHENYL-2,3-DIHYDRO-AS-TRIAZINE-3 –THIONE
A spectrophotometric procedure for the determination of Nickel (??) based on the complex formation with 6-phenyl-2, 3-dihydro-as-triazine-3-thione (PDTT) was developed. A yellow compound forms and is easily extractable by chloroform at pH>6. The mole ratio between Nickel (??) and PDTT is 1:4. The absorbance of the complex obeys Beer's law over the concentration range of 1-7 ug of Ni (??) pe...
متن کاملSimplified manual micromethod for determination of serum triglycerides.
This rapid, simplified assay for serum triglycerides is based on the use of a more effective solvent extraction procedure, ethoxide transesterification, and color development with acetylacetone. No phospholipid adsorption step is necessary. The procedure requires less than 0.2 ml of serum, no expensive or elaborate apparatus, and 5 to 10 values can be obtained per hour, with better precision th...
متن کاملSPECTROPHOTOMETRIC REACTION RATE METHOD FOR THE DETERMINATION OF SLWR BASED ON ITS CATALYTIC EFFEICT ON THE OXIDATION OF GLLOCYANW BY PER0XYDISULPTE
A sensitive and selective procedure was developed for the spectrophotometric determination of silver by its catalytic effect on the oxidation of gallocyanine by peroxydisulphate in acidic media at 540 nm. Silver can be determined in the range of 0.001-0.500 µg/ml with a limit of detection of 0.0005 µg/ml, by a fixed-time method of 4.0 min from initiation of the reaction, The relative standa...
متن کاملAn improved extraction procedure for the determination of triglycerides and cholesterol in plasma or serum.
A simple method of preparing extracts of plasma or serum suitable for determination of both triglycerides and cholesterol is described. Its chief advantage over present procedures is in a considerable saving of time and labor involved in the preparation of large numbersof samples. The method appearsto give a more complete extraction of glyceridesand comparesfavorably with existing methods of ex...
