چکیده

Thank you for submitting your manuscript for consideration by the EMBO Journal. It has now been seen by two referees whose comments are shown below. We share the referees' interest in this manuscript, but also felt at the editorial level that the dataset could be developed realistically to allow more general as well as more mechanistically developed conclusions. I outline below key points raised by the referees. I apologize for the atypically slow reviewing process due to some delays at the referee and unfortunately also at the editorial level due to high submission volumes during the main conference and holiday season.

A DR4:tBID axis drives the p53 apoptotic response by promoting oligomerization of poised BAX

The cellular response to p53 activation varies greatly in a stimulus- and cell type-specific manner. Dissecting the molecular mechanisms defining these cell fate choices will assist the development of effective p53-based cancer therapies and also illuminate fundamental processes by which gene networks control cellular behaviour. Using an experimental system wherein stimulus-specific p53 respons...

Bax oligomerization in mitochondrial membranes requires tBid (caspase-8-cleaved Bid) and a mitochondrial protein.

In response to various apoptotic stimuli, Bax, a pro-apoptotic member of the Bcl-2 family, is oligomerized and permeabilizes the mitochondrial outer membrane to apoptogenic factors, including cytochrome c. Bax oligomerization can also be induced by incubating isolated mitochondria containing endogenous Bax with recombinant tBid (caspase-8-cleaved Bid) in vitro. The mechanism by which Bax oligom...

Synergistic Effect of Fluorouracil Combined With Harmin on Induction of Apoptosis in Aspc-1 Cell Line And The Expression of Apoptotic Genes (Bax, P53 And Caspase3 &9)

Introduction: Pancreatic cancer is one of the deadliest cancers of the digestive system in the world. Due to the increasing resistance of cancer cells to chemotherapy and its side effects, there is a need for drugs with fewer side effects and natural origin. Methods: In this study, the effect of harmine and its concomitant use with fluorouracil to induce apoptosis in pancreatic cancer cells, A...

Mechanistic Issues of the Interaction of the Hairpin-Forming Domain of tBid with Mitochondrial Cardiolipin

BACKGROUND The pro-apoptotic effector Bid induces mitochondrial apoptosis in synergy with Bax and Bak. In response to death receptors activation, Bid is cleaved by caspase-8 into its active form, tBid (truncated Bid), which then translocates to the mitochondria to trigger cytochrome c release and subsequent apoptosis. Accumulating evidence now indicate that the binding of tBid initiates an orde...

VDAC2 is required for truncated BID-induced mitochondrial apoptosis by recruiting BAK to the mitochondria.

Truncated BID (tBID), a proapoptotic BCL2 family protein, induces BAK/BAX-dependent release of cytochrome c and other mitochondrial intermembrane proteins to the cytosol to induce apoptosis. The voltage-dependent anion channels (VDACs) are the primary gates for solutes across the outer mitochondrial membrane (OMM); however, their role in apoptotic OMM permeabilization remains controversial. Her...
