چکیده

The cornified cell envelope structure is formed beneath the plasma membrane in terminally differentiating stratified squamous epithelia. It provides a vital physical barrier to these tissues in mammals and consists of a 10 nm thick layer of highly crosslinked insoluble proteins. In the specialized case of the epidermis, a 5 nm thick layer of ceramide lipids is covalently bound to the proteins. These organize extracellular lipids into orderly lamellae and, together, the cell envelope and extracellular lipids are essential for effective physical and water barrier function in the skin. The assembly of the cornified cell envelope proceeds in three principal stages, starting in the upper spinous cell layers of the epidermis.

Differentially expressed late constituents of the epidermal cornified envelope.

Barrier activity of skin and internal barrier-forming epithelial linings are conferred by a lipid-corneocyte structure (stratum corneum in skin). The integrity of the corneocytes depends on the outer cornified envelope and is essential for maintenance of barrier function. During epidermal development and differentiation, proteins are sequentially incorporated into the envelope via action of epi...

Synthetic epidermal pentapeptide and related growth regulatory peptides inhibit proliferation and enhance differentiation in primary and regenerating cultures of human epidermal keratinocytes.

A pentapeptide that inhibits proliferation of mouse epidermal keratinocytes in vivo and in vitro has been purified from mouse skin extracts. In the present study the effect of a synthetic analog of the epidermal pentapeptide on proliferation and differentiation of cultured human epidermal keratinocytes was investigated. In young, rapidly growing primary cultures the pentapeptide caused a dramat...

Differentiation of the epidermal keratinocyte in cell culture: formation of the cornified envelope.

Human epidermal keratinocytes grow from single cells into stratified colonies. Cells in the upper layers of the colonies lose their ability to divide and begin terminal differentiation. In this process, there develops a cornified cell envelope that remains insoluble after heating in solutions of sodium dodecylsulfate and beta-mercaptoethanol. The insolubility of the cornified envelope depends u...

Cathepsin D is involved in the regulation of transglutaminase 1 and epidermal differentiation.

We previously demonstrated that the aspartate protease cathepsin D is activated by ceramide derived from acid sphingomyelinase. Increased expression of cathepsin D in the skin has been reported in wound healing, psoriasis and skin tumors. We explored specific functions of cathepsin D during epidermal differentiation. Protein expression and enzymatic activity of cathepsin D increased in differen...

Mice deficient in involucrin, envoplakin, and periplakin have a defective epidermal barrier

The cornified envelope is assembled from transglutaminase cross-linked proteins and lipids in the outermost epidermal layers and is essential for skin barrier function. Involucrin, envoplakin, and periplakin form the protein scaffold on which the envelope assembles. To examine their combined function, we generated mice deficient in all three genes. The triple knockouts have delayed embryonic ba...
