Discrimination of Normal and Abnormal Prothrombin and Protein C in Plasma Using a Calcium ton-Inhibited Monoclonal Antibody to a Common Epitope on Several Vitamin K-Dependent Proteins

چکیده
Vitamin K deficiency or administration of vitamin K antagonists results in the biosynthesis of abnormal des-’y-carboxy forms of the vitamin K-dependent proteins. Monoclonal antibody H-i 1 binds several vitamin K-dependent proteins at a determinant that includes the first two residues of ‘y-carboxyglutamic acid. Antibody H-i 1 binds fully carboxylated prothrombin and protein C in the presence of EDTA but binding is inhibited by the divalent metal ions. calcium. magnesium. and manganese. By contrast. des-y-carboxy prothrombin and protein C bind antibody H-i i the same in the presence of EDTA or calcium ion. Antibody H-i i thus appears to bind a conserved antigenic site containing y-carboxyglutamic acid that in the presence of divalent metal ion undergoes a conformational transition. This ability of antibody H-i 1 to bind des-’y-carboxy prothrombin

متن کاملProduction and Characterization of Murine Monoclonal Antibodies Recognizing Conformational and Linear Epitopes Localized on Human IgA2 Molecules
Background: There are two subclasses of human IgA (IgA1 and IgA2) that differ in antigenic properties and in chemical composition. The constant domains of α1 and α2 heavy chains have >95% sequence homology though major structural differences exist in the hinge region. Quantitation of IgA subclass levels depends on the availability of monoclonal antibodies (MAbs) specific for conserved conformat...
متن کاملAntibody Response to Human Extracellular HER2 Subdomain Proteins in Mice
Background: In addition to passive immunotherapy using anti-HER2 monoclonal antibodies, active immunotherapy via HER2 targeting is an interesting approach to inducing specific anti-tumor immune responses. We have recently reported the immunogenicity of HER2 subdomains following DNA immunization and HER2 protein boosting. In the present study, we evaluated the immunogenicity of different HER2 ex...
متن کاملMONOCLONAL ANTIBODIES TO HUMAN ALPHA-FETOPROTEIN
Alpha-fetoprotein (AFP), a serum glycoprotein belonging to the onco-developmental proteins group, serves as a marker both in cancer research and in studies concerning fetal development and fetal pathophysiology. Monoclonal anti-AFP antibodies are essential reagents in developing appropriate techniques for measurement of this protein. In this study, in order to produce anti-AFP monoclonal a...
متن کاملProduction of Monoclonal Antibody against Prokaryotically Expressed G1 Protein of Bovine Ephemeral Fever Virus
Epitope-G1 of bovine ephemeral fever virus (BEFV) G glycoprotein has been genetically and antigenically conserved among various isolates of BEFV and only reacts with anti-BEFV neutralising antibodies. Therefore, it is a candidate antigen for development of the enzyme linked immunosorbent assay (ELISA) for serological identification bovine ephemeral fever (BEF)-infected animals. The aim of this ...
