چکیده

The effect of 1,3,5(i 0)-estratriene-3,1 6a,i 7f@-triol(estriol), i ,3,5(i O)-estratriene-2,3-diol-i 7-one (2-hydroxyestrone), and 1,3,5(i 0)-estratriene-2,3,i 7$-triol (2-hydroxyestradiol) on the growth of dirnethylbenz(a)anthracene-induced mammary tumor and of R3230AC-transplantable mammary tumor was corn pared with that produced by estradiol benzoate treatment. Estriol showed minimal inhibition of tumor growth in dirnethyl benz(a)anthracene-induced tumor and no effect on R3230AC tumor while 2-hydroxyestrone showed no effect of tumor inhi bition. On the other hand, 2-hydroxyestradiol showed appre ciable inhibition of tumor growth in both tumors studied. That 2-hydroxyestradiol has been found to bind to estrogen recep tors in mammary tumors and is uterotropic suggests that the inhibition of tumor growth by 2-hydroxyestradiol may be similar to the mechanism of inhibition of mammary tumors by high concentrations of estradiol.

Phytochemicals inhibit catechol-O-methyltransferase activity in cytosolic fractions from healthy human mammary tissues: implications for catechol estrogen-induced DNA damage.

Phytochemicals are natural dietary constituents of fruits and vegetables. Some of these phytochemicals are known to affect estrogen-metabolizing enzymes. In breast tissue, estradiol can be metabolized to the catechol estrogens 2- and 4-hydroxyestradiol (2-OHE(2) and 4-OHE(2)). Catechol estrogens are suspected carcinogens potentially involved in the etiology of breast cancer. Catechol-O-methyltr...

Androgen Effects Mediated by Estrogen Receptor in 7,12- Dimethylbenz(a)anthracene-induced Rat Mammary Tumors1

To improve our understanding of the effects of androgens on the induction and growth of mammary tumors, the interaction of androgen on the estrogen receptor (ER) has been evaluated in the 7,12-dimethylbenz(a)anthracene-induced rat mammary tumor. In vitro competitive experiments have shown that androgens interact on ER with a low affinity but have a good stereospecificity for C,9 steroids bearin...

Is 2-methoxyestradiol an endogenous estrogen metabolite that inhibits mammary carcinogenesis?

Catechol estrogens (2- or 4-hydroxyestradiol and 2- or 4-hydroxyestrone) are chemically reactive estrogen metabolites that are O-methylated to less polar monomethyl ethers by catechol-O-methyltransferase, an enzyme present in many tissues such as the liver, kidney, brain, placenta, uterus, and mammary gland. In the present report, we review recent studies on the antitumorigenic and antiangiogen...

Effects of two new dihydropyridine derivatives, mepudipine and dibudipine, on isolated human internal mammary artery and rat left atrium

Among the present classes of calcium channel blockers, dihydropyridine derivatives are widely used in the therapy of hypertension, angina pectoris and the other cardiovascular diseases. Since the prototype of dihydropyridine derivatives, nifedipine, does not have the optimum pharmacokinetic and pharmacodynamic characteristics, several attempts have been made to synthesize other drugs in this ...
