چکیده

Carbohydrate starvation is an efficient stress treatment for induction of microspore embryogenesis. Transcriptome analysis of anthers response to mannitol treatment using the 22k Barley1 GeneChip revealed large changes in gene expression. Statistical analysis and filtering for 4-fold or greater changes resulted in 2,673 genes, of which 887 were up-regulated and 1,786 down-regulated. Great differences in some metabolic pathways, accompanied by a multi-dimensional stress response were found. Analysis of transcription factors showed that most of the down-regulated transcription factors were related to growth and development, and the up-regulated with abiotic and biotic stress responses and changes in developmental programs. Interestingly, the expression of most cell cycle related genes did not change significantly. Transcriptome analysis provided a successful approach to identify genes involved in mannitol treatment, essential for triggering microspore embryogenesis.

Expression profiles in barley microspore embryogenesis

In barley, microarray technology has allowed to study the gene expression profiling associated with the stress pretreatment phase of microspore embryogenesis, where the reprogramming of microspores takes place. Transcriptome analysis of anthers before and after four days of mannitol pretreatment, revealed changes in the expression of 2,673 genes. A thorough study of these differentially express...

Doubled haploid production is one of the best breeding methods to produce homozygous (pure) lines. Microspore culture technique is the most efficient method for production of doubled haploid plants. This research was carried out to study the effect of genotypes, stress pre-treatments and culture media on the callus induction and plant regeneration from microspores of anthers in barley (Hordeum ...

Efficient Microspore Isolation Methods for High Yield Embryoids and Regeneration in Rice (Oryza sativa L.)

Through anther and microspore culture methods, complete homozygous plants can be produced within a year as compared to the long inbreeding method. Isolated microspore culture is one of the most important techniques for rapid development of haploid plants. The efficiency of this method is influenced by several factors such as cultural conditions, growth regulators, plant media, pretreatments, ph...

Effects of Heat Shock and 2, 4-D Treatment on Morphological and Physiological Characteristics of Microspores and Microspore-Derived Doubled Haploid Plants in Brassica napus L.

Background:  Stresses such as heat shock, starvation, or osmotic is essential to lead isolated microspores towards embryogenesis. Despite the effectiveness of stresses in embryogenesis, they exert adverse effects on metabolism and growth of the regenerated plants. Objectives: The effects of heat shock and 2,4-D treatment on total protein content of treated microspores, morphological and physiol...

Production of haploid plants is an efficiant method in plant breeding programs, genetics and cytogenetics studies. Effects of cultivar, medium and plant growth regulators were investigated on anther culture of strawberry. This study was carried out using two commercial cultivars of satrawberry (Selva and Paros), three medium (MS, H1 and N6) and different concentrations of IAA and BAP as factori...
