چکیده

Armstrong, B. A. (The University of Kansas, Lawrence), and C. P. Sword. Electron microscopy of Listeria monocytogenes-infected mouse spleen. J. Bacteriol. 91:1346-1355. 1966.-Mouse spleen infected with Listeria monocytogenes was observed during the acute phase of infection; 72 hr after infection, organisms were usually found within phagocytic vacuoles in the cytoplasm of macrophages. These vacuoles, which resembled phagosomes, often contained several organisms as well as varying amounts of amorphous electron-dense material, ferritin-like particles, membrane fragments, and vesicles of varying density. Breakdown of vacuolar membranes appeared to be accompanied by damage to the host cell cytoplasm. Nuclear membrane damage was occasionally observed when phagocytic vacuoles were close to the nucleus.

Class Ia MHC-deficient BALB/c mice generate CD8+ T cell-mediated protective immunity against Listeria monocytogenes infection.

CD8(+) T cells are required for protective immunity against intracellular pathogens such as Listeria monocytogenes. In this study, we used class Ia MHC-deficient mice, which have a severe reduction in circulating CD8(+) T cells, to determine the protective capacity of class Ib MHC-restricted T cells during L. monocytogenes infection. The K(b-/-)D(b-/-) mutation was backcrossed onto a C.B10 (BAL...

Survival of Listeria monocytogenes in experimentally infected mice.

Physiological saline was found to be very detrimental to the viability of Listeria monocytogenes. The LD(50) value was substantially reduced when peptone was used as the suspending fluid rather than saline. Normal splenic tissue was not inhibitory to the survival of Listeria. In experimentally infected mice, L. monocytogenes survived for 8 days in the peritoneal cavity and for at least 11 days ...

Mechanism of depletion of T lymphocytes from the spleen of mice infected with Listeria monocytogenes.

Marked changes in the splenic lymphocyte populations during murine infection with Listeria monocytogenes were observed histologically and quantitated by the immunofluorescence of Thy-1+ immunoglobulin (Ig-) (T) and Ig+ (B) cells. Cells were depleted from the T-dependent areas of the spleen, and the number of T cells in suspensions prepared from spleens of mice 1 to 3 days after primary or secon...

Listeria monocytogenes-reactive T lymphocyte clones with cytolytic activity against infected target cells

Lyt-2+ T cell clones were established from Listeria monocytogenes-infected mice. The clones secreted IFN-gamma after stimulation with spleen cells from L. monocytogenes-infected mice plus IL-2. Spleen cells from normal mice were not stimulatory. Furthermore, cloned T cells lysed L. monocytogenes-infected macrophages. Cytolysis was antigen-specific and H-2K-restricted. These findings suggest a r...

Bacterial entry to the splenic white pulp initiates antigen presentation to CD8+ T cells.

The spleen plays an important role in host-protective responses to bacteria. However, the cellular dynamics that lead to pathogen-specific immunity remain poorly understood. Here we examined Listeria monocytogenes (Lm) infection in the mouse spleen via in situ fluorescence microscopy. We found that the redistribution of Lm from the marginal zone (MZ) to the periarteriolar lymphoid sheath (PALS)...
