Regulation of Cell-cell Adhesion and Actin Cytoskeleton in Non-transformed and Transformed Epithelial Cells

چکیده
Epithelial cell-cell adhesions have a critical role in morphogenesis, establishment and maintenance of tissue architecture, cell-cell communication, normal cell growth and differentiation. These adhesions are disrupted during malignant transformation and tumour cell invasion. Several kinases, phosphatases and small GTPases regulate cell-cell contacts. In the present work we investigated the dynamics of cell-cell adhesion structures after microinjection of fluorophore tagged vinculin, during transformation caused by an active Src tyrosine kinase and during Helicobacter pylori infection. The regulatory role of Rac GTPase as well as the behaviour of actin and cadherin were analysed in all

Rab11 is required for maintenance of cell shape via βPS integrin mediated cell adhesion in Drosophila
In eukaryotes, vesicle trafficking is regulated by the small monomeric GTPases of the Rab protein family. Rab11, (a subfamily of the Ypt/Rab gene family) an evolutionarily conserved, ubiquitously expressed subfamily of small monomeric Rab GTPases, has been implicated in regulating vesicular trafficking through the recycling of endosomal compartment. In an earlier communication, we have shown th...

E-cadherin-mediated cell-cell adhesion, which is essential for the maintenance of the architecture and integrity of epithelial tissues, is often lost during carcinoma progression. To better understand the nature of alterations of cell-cell interactions at the early stages of neoplastic evolution of epithelial cells, we examined the line of nontransformed IAR-2 epithelial cells and their descend...
متن کاملInhibition of anchorage-independent growth of transformed NIH3T3 cells by epithelial protein lost in neoplasm (EPLIN) requires localization of EPLIN to actin cytoskeleton.
Epithelial protein lost in neoplasm (EPLIN) is a cytoskeleton-associated protein characterized by the presence of a single centrally located lin-11, isl-1, and mec-3 (LIM) domain. We have reported previously that EPLIN is down-regulated in transformed cells. In this study, we have investigated whether ectopic expression of EPLIN affects transformation. In untransformed NIH3T3 cells, retroviral-...
متن کاملO 7: KCNK2 Regulates the Nanoscale Formation of Immune Docking Structures on Brain Endothelial Cells Under Autoinflammatory Conditions
KCNK2 was previously shown to regulate immune-cell trafficking into the central nervous system (CNS). Kcnk2-/- mice demonstrated a more severe disease course in experimental autoimmune encephalomyelitis, an animal model of multiple sclerosis, due to an increased immune-cell migration into the CNS. An upregulation of the cellular adhesion molecules ICAM1 and VCAM1 on brain endothelial cells in K...
متن کاملInverse relationship between TCTP/RhoA and p53 /cyclin A/actin expression in ovarian cancer cells.
The translationally controlled tumor protein (TCTP) plays a role in cell growth, cell cycle and cancer progression. TCTP controls negatively the stability of the p53 tumor suppressor protein and interacts with the cellular cytoskeleton. The deregulation of the actin and cytokeratin cytoskeleton is responsible for the increased migratory activity of tumor cells and is linked with poor patient ou...
