CELL LINEAGE CHOICE DURING DIFFERENTIATION OF TROPHOBLAST STEMCELLS (TSC) IS DEPENDENT ON OXYGEN LEVELS, AND MEDIATED BY STRESSENZYME PATHWAYS AND MITOCHONDRIAL FUNCTIONbySICHANG ZHOUDecember 2011Advisor: Dr. Daniel A. RappoleeMajor: Physiology, Concentration in Reproductive SciencesDegree: Doctor of PhilosophyIn general, stress is defined as a noxious factor in a broad spectrum (chemical, biological or physical). Stress response of human body can be divided into three levels-integrate, organ/tissue or cellular response. Trophoblast stem cell (TSC) is one of theearliest differentiated stem cell within blastocyst, which is the stage of preimplantationembryo developed from fertilized egg. Different from another pluripotent embryonicstem cell, which is derived from inner cell mass in blastocyst and develop into fetus, multipotent TSC will specifically differentiate into all kinds of cell subtypes consisting ofplacenta by which nutrient and oxygen from mom as well as metabolites from fetus canbe exchanged. In our previous studies based upon preimplantation embryo and TSC, fourparameters for defining stress are identified: 1) decreased cell accumulation; 2)increased apoptosis; 3) decreased mutipotent markers and increased cell lineagemarkers; 4) increased activity of stress enzyme SAPK/JNK. By these parametersapplying on different O2 cultured TSC, we identified that 2% O2 is the optimal oxygenenvironment for TSC mutipotency maintenance and proliferation. Additionally, TSC

Hypoxic stress induces, but cannot sustain trophoblast stem cell differentiation to labyrinthine placenta due to mitochondrial insufficiency.

Dysfunctional stem cell differentiation into placental lineages is associated with gestational diseases. Of the differentiated lineages available to trophoblast stem cells (TSC), elevated O2 and mitochondrial function are necessary to placental lineages at the maternal-placental surface and important in the etiology of preeclampsia. TSC lineage imbalance leads to embryonic failure during uterin...

Cellular stress causes reversible, PRKAA1/2-, and proteasome-dependent ID2 protein loss in trophoblast stem cells.

Stress reduces fertility, but the mechanisms mediating this are not understood. For a successful pregnancy, placental trophoblast stem cells (TSCs) in the implanting embryo proliferate and then a subpopulation differentiates to produce hormones. Normally, differentiation occurs when inhibitor of differentiation 2 (ID2) protein is lost in human and mouse placental stem cells. We hypothesize that...

Nuclear Architecture and Epigenetics of Lineage Choice

Differentiation is an epigenetic process which is installed by changes of transcriptional programs over successive cellular divisions. A number of studies have reported the effects of biochemical modifications of chromatin (DNA and chromatin proteins) on the regulation of transcription. Although, these studies are able to explain how transcription of a given gene is regulated (toward activation...

Promoter Methylation and Gene Expression in Human CD34+ Stem Cells Derived Erythroid Lineage by MicroRNA

Background: Stem Cell differentiation is a process composed of vast variety of factors which are controlled by a network of certain mechanisms. This study aims to determine the possible role of DNA methylation, a potent regulator of VHL, ECAD and RUNX3 genes during Erythroid differentiation driven by miR-451. Materials and Methods: To determine the methylation status of promoters and the e...

ECM-Dependent HIF Induction Directs Trophoblast Stem Cell Fate via LIMK1-Mediated Cytoskeletal Rearrangement

The Hypoxia-inducible Factor (HIF) family of transcriptional regulators coordinates the expression of dozens of genes in response to oxygen deprivation. Mammalian development occurs in a hypoxic environment and HIF-null mice therefore die in utero due to multiple embryonic and placental defects. Mouse embryonic stem cells do not differentiate into placental cells; therefore, trophoblast stem ce...
