چکیده

BACKGROUND Bioactive molecules have received increasing attention due to their nutraceutical attributes and anticancer, antioxidant, antiproliferative and apoptosis-inducing properties. This study aimed to investigate the biological properties of carotenoids extracted from Archaea. METHODS Halophilic Archaea strains were isolated from the brine of a local crystallizer pond (TS7) of a solar saltern at Sfax, Tunisia. The most carotenoid-producing strain (M8) was investigated on heptoma cell line (HepG2), and its viability was assessed by the MTT-test. The cells were incubated with different sub-lethal extract rates, with carotenoid concentrations ranging from 0.2 to 1.5 μM. Antioxidant activity was evaluated through exposing the cells to sub-lethal extract concentrations for 24 hours and then to oxidative stress induced by 60 μM arachidonic acid and 50 μM H₂O₂. RESULTS Compared to non-treated cells, bacterial carotenoid extracts inhibited HepG2 cell viability (50%). A time and dose effect was observed, with cell viability undergoing a significant (P < 0.05) decrease with extract concentration. After exposure to oxidative stress, control cells underwent a significant (P < 0.05) decrease in viability as compared to the non-treated cells. CONCLUSIONS The bacterial extracts under investigation were noted to exhibit the strongest free radical scavenging activity with high carotenoid concentrations. The carotenoid extract also showed significant antiproliferative activity against HepG2 human cancer cell lines.

Ch2, a novel halophilic archaeon from an Australian solar saltern.

A novel halophilic archaeon, strain Ch2, was isolated from a marine solar saltern in Geelong, Australia. The fact that this organism had a dam-methylated genome suggested that it is closely related to the taxon that includes Halobacterium saccharovorum, Halobacterium sodomense, and Halobacterium trapanicum. A sequence analysis of the 16S rRNA gene (Ch2 has three copies of this gene) showed that...

Isolation of Photochrome Transmembrane Protein Bacteriorhodopsin from Purple Membranes of Halobacterium Halobacterium Halobium. New Method for Isolation

This paper presents improved method for isolation of photochrome transmembraine protein bacteriorhodopsin (output 5 mg from 100 g of wet biomass) capable to transform light energy to electrochemical energy of generated protons H + and АТP. The protein was isolated from purple membranes of photo-organotrophic halobacterium Halobacterium halobium by cellular autolysis by distilled water, processi...

Spectrophotometric identification of the pigment associated with light-driven primary sodium translocation in Halobacterium halobium.

It has been recently proposed that in addition to bacteriorhodopsin, Halobacterium halobium membranes contain a second light-reactive pigment, whose function is the translocation of sodium ions. The evidence is based on the presence of light-driven, uncoupler-insensitive, sodium ion extrusion and light-driven, uncoupler-enhanced, passive proton uptake in cell envelope vesicles prepared from H....

Sensory transduction in Halobacterium halobium: retinal protein pigment controls UV-induced behavioral response.

Both photosystems, PS 370 and PS 565, controlling behavioral responses in Halobacterium halobium [E. Hildebrand and N. Dencher, Nature 2 5 7 ,4 6 — 48 (1975)] are reversibly inhibited when bacteria are grown in the presence o f 1 m M nicotine which is known to block biosynthesis o f reti nal. Photobehavior can be restored within some minutes to hours by adding retinal to nicotinetreated bacter...

Common structural features of the genes for two stable RNAs from Halobacterium halobium

The genes coding for the 5S rRNA and another stable RNA, termed 7S RNA, in Halobacterium halobium were isolated from a genomic library of this archaebacterium and their nucleotide sequences determined. Both genes are colinear with their transcripts (5S rRNA and 7S RNA), but 5S rRNA and possibly also 7S RNA isolated from other halobacteria carry additional nucleotides within the RNA transcript. ...
