چکیده

The effects of formycin (7-amino-3-(/3-D-ribofuranosyl)pyrazolo(4,3-d)-pyrimidine), an adenosine analog, on purine metabolism in Ehrlich ascites tumor cells were studied. Purine biosynthesis de novo was blocked by formycin through inhibition of phosphoribosylpyrophosphate synthesis, and this did not occur in cells which did not phosphorylate formycin. This analog had no significant effect on adenine conversion to adenine and guanine nucleotides, but did inhibit the synthesis of these nucleotides from hypoxanthine and guanine. The incorporation of purines into nucleic acids was inhibited slightly. Formycin did not inhibit the phosphorolysis of guanosine or inosine, but did inhibit the incorporation of lysine and methionine into protein. For mycin 5'-phosphate inhibited adenine phosphoribosyltransferase

Biochemical effects of formycin B on Xanthomonas oryzae.

Formycin B, an inosine analogue, inhibits the incorporation of radioactive adenosine, uridine and thymidine, but none of formic acid, orotic acid and inorganic phosphoric acid into the cellular nucleic acid of Xanthomonas oryzae. The biochemical effects are readily reversed when formycin B is removed. Cell-free extract from the organism is entirely insensitive to formycin B in respect to phosph...

Biochemical studies of the nucleoside analogue, formycin.

The absorption and fluorescence properties of formycin provide a useful tool in the analysis of enzyme-substrate interactions; we have therefore studied selected biochemical properties of formycin nucleotides. Formycin and its derivatives effectively substitute for the corresponding adenosine substrates in a wide variety of enzyme reactions. These include enzymes of nucleotide metabolism, RNA p...

Effects of a New Adenosine Deaminase Inhibitor, Isocoformycin, on Toxicity, Antitumor Activity and Tissue Distribution of Formycin a and 9-b-d-arabinofuranosyladenine

Single intraperitoneal and intravenous injections of isocoformycin at 1,200 mg/kg did not cause the death of mice. Isocoformycin which inhibited adenosine deaminase enhanced significantly the toxicity of formycin A and ara-A at various combination ratios. Isocoformycin potentiated antitumor activity of formycin A and ara-A against L1210 leukemia. Formycin A and ara-A disappeared rapidly from th...

Leishmaniasis is initiated when sandflies inject the extracellular promastigote form of the parasite into the skin. Promastigotes are rapidly phagocytized, after which they transform into the intracellular amastigote form. The multiplication of the latter within macrophages leads to clinical disease. Treatment of leishmaniasis is primarily with pentavalent antimonials, parenteral and potentiall...

Metabolism of tubercidin and formycin to their 3':5'-cyclic nucleotides in mammalian cells.

High performance anion exchange chromatography was utilized to demonstrate extensive metabolism of tubercidin and formycin to their corresponding 5’-triphosphates in mouse leukemia EL4 cells and a concomitant decrease in cellular ATP. Efficient anabolism of formycin required the presence of an inhibitor (erythro9-(2-hydroxy-3-nonyl)adenine) of adenosine deaminase in the cellular incubations. Ac...
